针灸对CTX化疗荷瘤小鼠脾脏组织中IL-12和TNF-α含量的影响 被引量:12

Effects of acupuncture-moxibustion on contents of IL-12 and TNF-αin spleen of cyclophosphamideinduced cancer-bearing mice

摘要 目的:探讨针灸改善荷瘤小鼠化疗所致免疫功能损伤的分子生物学机制。方法:将72只荷瘤模型建立成功小鼠,采用随机分层分组法,3天批、5天批各36只,均随机分为空白组、模型组、针刺组、艾灸组,每组9只。除空白组外,余组小鼠一次性腹腔注射环磷酰胺(cyclophosphamide,CTX)溶液(150mg/kg),制备CTX荷瘤小鼠模型;空白组腹腔注射同等剂量的0.9%NaCl溶液。4h后针刺组、艾灸组分别予以针刺、艾灸治疗,穴取"大椎""膈俞""肾俞""足三里",每天1次,空白组、模型组每日同时间抓取固定,不治疗。分别于治疗3天、5天后的第2天取材,采用ELISA法检测各组小鼠脾脏组织中白介素-12(IL-12)和肿瘤坏死因子-α(TNF-α)含量。结果:治疗3天、5天后,与空白组比较,模型组荷瘤小鼠脾脏组织中IL-12、TNF-α含量均降低(均P<0.05);与模型组比较,针刺组、艾灸组小鼠脾脏组织中IL-12、TNF-α含量均升高(均P<0.05);针刺组与艾灸组脾脏组织中IL-12、TNF-α含量比较差异均无统计学意义(均P>0.05)。结论:针刺和艾灸可以有效提高CTX化疗荷瘤小鼠脾脏组织中IL-12和TNF-α的含量,减轻化疗所致的免疫功能损伤,提高机体免疫功能。 Objective To explore the molecular biology mechanism of acupuncture on improving immune function damage induced by chemotherapy in cancer-bearing mice.Methods Seventy-two mice(36mice in 3-day treatment and 5-day treatment,respectively)which were successfully made into cancer-bearing model were divided into a blank group,a model group,an acupuncture group and a moxibustion group by stratified randomization method,9mice in each one.Except for the mice in the blank group,the remaining mice were treated with intra-peritoneal injection of cyclophosphamide(CTX,150mg/kg),to establish the cancer-bearing mice of CTX.The mice in the blank group were treated with intraperitoneal injection of 0.9% NaCl(identical dose as other groups).After 4h,the mice in the acupuncture group and moxibustion group were treated with acupuncture and moxibustion at"Dazhui"(GV 14),"Geshu"(BL 17),"Shenshu"(BL 23),"Zusanli"(ST 36),once a day.The mice in the blank group and model group were treated with immobilization and fixation during the same time.On the next day of the end of 3-day and 5-day treatment,the sample was collected.The ELISA method was applied to measure the contents of IL-12 and TNF-αin spleen of all the mice.Results After 3-day and 5-day treatment,compared with the blank group,the contents of IL-12 and TNF-αin spleen in the model group were reduced(all P0.05);compared with the model group,the contents of IL-12 and TNF-αin spleen in the acupuncture group and moxibustion group were increased(all P0.05),but the content of IL-12 and TNF-αin the acupuncture group was not different from that in the moxibustion group(both P0.05).Conclusion Acupuncture and moxibustion could effectively increase the contents of IL-12 and TNF-αin spleen of CTX cancer-bearing mice,which could relieve chemotherapy-induced immune function damage to improve immune function.
