摘要
目的探讨玛卡中多糖的提取、纯化和含量测定的紫外分光光度方法。方法采用水提醇沉再经过碱性硫酸铜纯化的方法进行制备,在强酸性条件下多糖水解成单糖分子,在沸水浴中脱水生成羟甲基呋喃甲醛(羟甲基糖醛)再与苯酚缩合成淡橙黄色的化合物,其显色强度与溶液中单糖的浓度成正比,灵敏而稳定,比色测定溶液在485 nm波长处有最大吸收。结果对照品葡聚糖计的检测浓度在0.005 mg/ml^0.100 mg/ml时与吸光度呈良好的线性关系,得回归方程y=10.612 3x-0.001 7,r=0.999 6,平均回收率为96.89%,RSD为1.44%。结论紫外分光光度法灵敏、快速、准确、成本低,适用于玛卡中多糖含量测定。
Objective To investigate the method for extraction,purification and content determination of polysaccharides in Maca by ultraviolet spectrophotometric method. Methods The sample was processed by water extraction and alcohol precipitation and then by the purification of alkaline copper sulfate method. Polysaccharides,under stron g acidic conditions,were hydrolyzed into monosaccharide molecules. Followin g dehydration in a boilin g water bath,hydroxymethyl furaldehyde was produced,which was subjected to condensation with phenol to produce a pale orange yellow compound. The color intensity was in a sensitive and stable positive relationship with the concentration of monosaccharides in the solution and exhibited maximum light absorption at 485 nm wavelength in colorimetric determination. Results Good linearity was presented between the absorbance value and the concentration of the reference substance( poly glucosan) over the range of 0. 005 mg / ml-0. 100 mg / ml. The regression equation was y = 10. 612 3x-0. 001 7( r = 0. 999 6). The average recovery rate was 96. 89% and the RSD was1. 44%. Conclusion Ultraviolet spectrophotometry is sensitive,rapid,stable and cost-effective and suitable for determination of polysaccharides in Maca.
