摘要
目的比较3种方法检测抗ds-DNA抗体的阳性率及临床实用性,为临床中小型实验室选择合适的临床检测方法提供准确可靠的依据。方法对90例系统性红斑狼疮(SLE)患者同时进行以绿蝇短膜虫为基质抗原的间接免疫荧光法(IIFA)、免疫印迹法(IBT)和酶联免疫法(ELISA)检测,检测结果以间接免疫荧光法为参考方法进行比较分析。结果IIFA、IBT、ELISA检测方法的阳性率分别为77.8%、84.4%、82.2%。以IIFA为参考方法算得IBT、ELISA与IIFA的阳性符合率分别是95.7%、97.1%。结论免疫印迹法、酶联免疫法与间接免疫荧光法的阳性符合率均在95%以上,此2种方法无需荧光显微镜,且结果更易判读,因此对于没有荧光显微镜的临床中小型实验室,同样可以选择免疫印迹法或酶联免疫法进行抗ds DNA抗体的检测,为临床提供准确可靠的结果。
Objective Compare the positive rate and the clinical practical application with three detection methods of detecting anti ds-DNA antibodies,to provide accurate and reliable basis for selecting appropriate clinical detection methods for small and medium sized laboratories. Methods 90 cases of patients with systemic lupus erythematosus( SLE) were detected simultaneously by indirect immunofluorescence method( IIFA) with green fly short film insect as stromal antigen and western blot method( IBT) and enzyme-linked immunoassay( ELISA),the detection results were compared and analyzed by indirect immunofluorescence method as the reference method. Result The positive rate of IIFA was 77. 8%,the positive rate of IBT was 84. 4%and the positive rate of ELISA was 82. 2%. Using the IIFA for reference method,the positive coincidence rate of IBT,ELISA with IIFA was 95. 7%,97. 1% respectively. Conclusion The positive coincidence rate of western blot method,enzyme-linked immunoassay with indirect immunofluorescence were both over 95%,the two methods were conducted without fluorescence microscope,and the results are more likely to interpretation,so clinical small laboratory without fluorescence microscope can also choose western blot method or enzyme-linked immunoassay for detection of anti ds-DNA antibodies,to provide an accurate and reliable results for clinic.
