摘要
目的通过给予大鼠不同的慢性应激,探讨慢性应激对大鼠实验性牙周炎发病过程的影响。方法Wistar大鼠32只,随机分成空白牙周炎组(N组)、慢性应激牙周炎组(C组)2组,慢性应激6周后,每组的右侧下颌第一磨牙做实验性牙周炎模型,左侧做自身对照。观察测量2组的体重和平均牙周探诊深度,分离牙龈组织行HE染色观察组织学变化;免疫组化染色检测MMP-9,心脏取血,酶联免疫吸附法检测CTX-1并进行统计学分析。结果 C组与N组比较,组织学表现炎症明显,其体重、平均牙周探诊深度、MMP-9和CTX-1有显著统计学差异。结论慢性应激可以减缓大鼠体重的增长;可加重大鼠实验性牙周炎的病损。
Objective To observe the onset process of experimental periodontitis in rats, through establishment of chronic stress model. Methods 32 Wister rat,fallen into 2 groups randomly, normal periodontitis group(N group),periodontitis group impacted by chronic stress(C group).The C group was given different chronic stress,6 weeks later,mandibular first molar were made as experimental periodontitis model in right, and the left one as its own control.Observation the weight and mean periodontal probing depth in 2 groups; separated gingival tissue, HE staining, Sirius red staining, observed pathological changes;Immunohistochemical staining, determined MMP-9 levels, drawed blood from heart, used enzyme linked immunosorbent assay to detection CTX-1 levels. Results Compared with C group and N group, there were significant difference in weight, MMP-9, CTX-1 and mean periodontal probing depth. Conclusion Chronic stress could slow down the weight of rats growth;it could aggravate periodontal lesion degree in experimental periodontitis- rats.
