酮体生成和利用定量实验方法的建立

ESTABLISHMENT OF METHOD FOR QUANTITATIVE EXPERIMENT ON KETONE BODIES FORMATION AND UTILIZATION

摘要 以丁酸钠为作用物,利用组织切片培养法,建立了定量测定酮体生成和利用的实验方法。实验的最适条件是:大鼠饥饿48小时;气温10~20℃,以15℃为宜;丁酸钠浓度0.75mol/L;37℃恒温振荡培养时间4小时。依此,可定量测定肝脏生成或其它组织利用的酮体量。 An experimental method for quantitative determination of the ketonebodies formation and utilization has been established, by using sodiumbutyrate as substrate and applying the method of tissue slices culture. Optimalcondition are as follows: rat starved for 48h; atmospheric temperature 10~20℃,preferably 15℃; concentration of sodium butyrate 0.75mol/L; duration of tissueslices culture for 4h. According to above conditions, amount of ketonebodies formation in liver and utilization in other organs can be determinedquantitatively.
