益精方对腺嘌呤法大鼠睾丸组织中TGF-β3 mRNA及蛋白表达影响的实验研究 被引量:5

摘要 目的 观察益精方对腺嘌呤法不育症大鼠睾丸组织中TGF-β 3 mRNA及蛋白表达水平的影响.方法选用48只健康的SPF级Wistar雄性大鼠,2月龄,体质量(180·20)g,随机分为空白组、模型组、复方玄驹组和益精方组,每组12只.除空白组外,其余各组均以腺嘌呤1 mL/ (100g·d)的剂量连续灌胃12d;从第13天开始,空白组及模型组用等量生理盐水灌胃,复方玄驹组以复方玄驹胶囊水溶液灌胃,益精方组以益精方汤剂灌胃,每天一次,连续20d.实验结束后处死大鼠,摘取睾丸,采用免疫组化技术测定大鼠睾丸组织中TGF-β 3的表达;采用RT-PCR技术测定TGF-3 3 mRNA的表达.结果 与空白组比较,模型组中TGF-β 3及TGF-β 3 mRNA的表达均升高,差异均具有统计学意义(P<0.01).与模型组比较,复方玄驹组和益精方组中TGF-β 3及TGF-β 3 mRNA的表达均有所下降,差异均具有统计学意义(P<0.05).结论 益精方对腺嘌呤法诱导的大鼠睾丸组织中TGF-β 3及TGF-β3mRNA的高表达具有明显的抑制作用. Objective To observe the influence of Yijing Fang on the expression of TGF-beta 3 in the testis tissues of adenine -induced infertility rots. Methods Total of 48 SPF- grade healthy male Wistar rats with March age, weight (180~20)g, were randomly divided into four groups such as the blank group, the model group, compound Xuanju group, Y1jing Fang group, 12 rats in each group. Except rats in the blank group, rats in the other groups were intragastrically administratedwith a dose of 1 mL/(100g d)adenine for 12 days; On the 13th day, rats in the blank group and the model group were intragastrically treated with normal saline, and the rats in compound Xuanju group with compound Xuanju capsule aqueous garage, and the rats in Yijing Fang Group with Yijing Fang Decoction, once a day for 20 consecutive days. After the end of the experiment the rats were sacrificed and their testis and epididymis were collected for TGF- beta 3 expression analysis. Results Compared with that of the blank group, the expression of TGF- beta 3 model group was remarkably increased, the difference was statistically significant (P〈0.01). Compared with that of the model group, the expressions of TGF- beta 3 in compound Xuaaju group and Yi Jing Fang Group were all decreased, the difference was statistically significant (P〈0.05). Conclusion YijingFang may obviously inhibit high expression of TGF- beta 3 in testis of adenine-induced infertility rat.
