摘要
目的研究葛根复方(葛根、黄芪、丹参、苍术、黄柏等)对实验性自身免疫性重症肌无力大鼠腓肠肌重复性神经刺激电位(RNS)和乙酰胆碱受体抗体(AchR-Ab)表达的影响,并探讨其机制。方法将100只Lewis大鼠随机分为4组,除正常组,模型组、强的松组、葛根复方组采用皮下注射鼠源性AchR-α亚基97~116肽段方法建立重症肌无力的实验模型,造模成功的两组分别给予强的松、葛根复方灌胃4周,观察大鼠治疗前后的RNS和AchR-Ab的变化。结果治疗前与正常组比较,造模各组大鼠的RNS都有明显衰减;治疗后与模型组比较,强的松组和葛根复方组的RNS衰减程度都减弱;治疗后与强的松组比较,葛根复方组的RNS衰减程度明显低。治疗前造模各组大鼠的AchR-Ab表达水平明显高于正常组(P〈0.01);强的松组和葛根复方组治疗后和模型组比较,两组中AchR-Ab表达水平明显下降(P〈0.05);治疗后强的松和葛根复方组比较,葛根复方组的AchR-Ab表达水平明显偏低(P〈0.05)。结论葛根复方能促进实验性自身免疫性重症肌无力大鼠腓肠肌的肌力恢复。
AIM To research the effect of Compound Gegen Decoction( Puerariae lobatae Radix,Astragali Radix,Salviae miltiorrhizae Radix et Rhizoma,Atractylodis Rhizoma,Phellodendri chinensis Cortex,etc.) on gastrocnemius repetitive nerve stimulation( RNS) and the expression of acetylcholinereceptor antibodies( AchR-Ab)in rat serum with experimental autoimmune myasthenia gravis( EAMG) and its mechanism of action. METHODS One hundred Lewis rats were randomly and equally divided into four groups,except the control group,rats in other three groups,including the model,Compound Gegen Decoction and Prednisone groups,were immunized with rat-derived 97- 116 peptide of the AchRα-subunit. After the EAMG model was successfully made,the rats in two groups were administered with Prednisone and Compound Gegen Decoction for four weeks. The changes of RNS and AchR-Ab were observed in each group before and after treatment. RESULTS As compared with the control group before the treatment,RNS level in model rats decreased more. As compared with the model group after the treatment,RNS recession level in Prednisone and Compound Gegen Decoction groups showed more recoveries,but the latter was inferior to the former. As compared with the control group before the treatment,AchR-Ab level in model rats was obviously higher( P〈0. 01). After the treatment,AchR-Ab level in Compound Gegen Decoction and Prescription groups showed markedly lower( P〈0. 05),but the former was less than the latter( P〈0. 05).CONCLUSION Compound Gegen Decoction can promote the improvement of muscle strength in rats withEAMG.
