多成分综合评分法优化高良姜乙醇提取工艺研究 被引量:7

摘要 目的优化高良姜的乙醇提取工艺。方法建立高效液相色谱法,同时测定高良姜中5-羟基-7-(4-羟基-3-甲氧基苯基)-1-苯基-3-庚酮、高良姜素、山柰素、7-(4-羟基-3-甲氧基苯基)-1-苯基-4-烯-3-庚酮和1,7-二苯基-5-羟基-3-庚酮5种成分,采用L9(34)正交试验法,以上述5种成分含有量的综合评分为指标,考察乙醇体积分数、提取时间、提取次数和料液比4个影响因素,优选高良姜的醇提工艺。结果提取次数对高良姜的提取工艺有显著性影响,优选出的提取工艺为加10倍量80%乙醇,提取3次,每次1 h;高良姜中5种成分的转移率分别为5-羟基-7-(4-羟基-3-甲氧基苯基)-1-苯基-3-庚酮91.7%、高良姜素92.3%、山柰素85.0%、7-(4-羟基-3-甲氧基苯基)-1-苯基-4-烯-3-庚酮94.8%、1,7-二苯基-5-羟基-3-庚酮93.9%。结论多成分综合评分法优选出的提取工艺能对高良姜药材进行较完全地提取,可用于在线监控。 AIM To optimize the ethanol extraction of Alpinia officinarum. METHOD A high-performance liquid chromatography( HPLC) method was established to simultaneously determine the contents of 5-hydroxy-7-( 4-hydroxy-3-methoxy phenyl)-1-phenyl-3-heptanone,galangin,kaempferide,7-( 4-hydroxy-3-methoxy phenyl)-1-phenyl-4-ene-3-heptanone and 1,7-diphenyl-5-hydroxy-3-heptanone. With the comprehensive score of the conterts of five components as an indicator,ethanol concentration,extraction time,extraction frequency and the ratio of material to solvent were optimized by L9( 3^4) orthogonal design. RESULTS The extraction frequency played a key role in extraction,and the optimal conditions were one hour extraction at 80% ethanol concentration for three times and 1 ∶ 10 ratio of material to solvent. The extraction rates of 5-hydroxy-7-( 4-hydroxy-3-methoxy phenyl)-1-phenyl-3-heptanone,galangin,kaempferide,7-( 4-hydroxy-3-methoxy phenyl)-1-phenyl-4-ene-3-heptanone and 1,7-diphenyl-5-hydroxy-3-heptanone were 91. 7%,92. 3%,85. 0%,94. 8%,and 93. 9%,respectively. CONCLUSION The multiple components evaluation can help to make a complete extraction,which can be applied as an online monitor for Alpinia Officinarum extraction.
