冠盖藤相伍土鳖虫治疗膝骨性关节炎血瘀作用机制的研究 被引量:4

Action mechanisms of Pileostegia viburnoides combined with Eupolyphaga sinensis in treatment of knee osteoarthritis with blood stasis

摘要 目的:观察冠盖藤相伍土鳖虫对急性血瘀模型大鼠血液流变学以及对膝骨性关节炎模型大鼠纤溶系统t PA、PAI活性的影响,从而探讨其活血化瘀功效的作用机制。方法:用皮下注射大剂量盐酸肾上腺素和冰水刺激制备急性血瘀模型,检测主要血液流变学指标;用4%木瓜蛋白酶溶液与0.3 mol/L半胱氨酸溶液的混合液注入大鼠右膝关节制备膝骨性关节炎模型,检测血浆中t PA、PAI含量变化。结果:用药各组均能不同程度地改善急性血瘀模型大鼠血液流变学指标,相伍用药组优于冠盖藤单独用药组。各用药组与模型组比较都可以使血浆中t PA含量增多,PAI含量减少,相伍用药组明显优于冠盖藤单独用药组。两药相伍能更好地改善血液流变学及纤溶活性,对大鼠在膝骨性关节炎模型中所引起的纤溶系统活性的下降起到一定程度的抑制作用。结论:冠盖藤提取物与土鳖虫提取物相伍有改善血液流变学的作用,通过调节血浆t PA,PAI水平,达到改善骨性关节炎的血瘀症状。 Objective: To observe the influence of Pileostegia viburnoides extract combined with Eupolyphaga sinensis extract on hemorheological parameters in a rat model of acute blood stasis and the fibrinolytic activities of tissue plasminogen activator( t PA) and its fast acting inhibitor( PAI) in a rat model of knee osteoarthritis( KOA)and to explore their action mechanisms in activating blood circulation and resolving stasis. Methods: The rat model of acute blood stasis was established by subcutaneous injection of large doses of epinephrine hydrochloride and ice water stimulation. The main hemorheological parameters in this model were examined. The rat model of KOA was established by injection of a mixture of 4% papain solution and 0. 3 mol / L L- cysteine solution into the rat right knee. The changes in plasma t PA and PAI levels in this model were measured. Results: All treatment groups showed varying degrees of improvement in hemorheological parameters in the rat model of acute blood stasis. The synergy group showed greater improvement than the simple recipe( Pileostegia viburnoides) group. All treatment groups and model groups had an increase in the t PA content and a reduction in the PAI content. The synergy group had greater changes in these parameters than the simple recipe( Pileostegia viburnoides) group. Conclusion: A combination of Pileostegia viburnoides extract and Eupolyphaga sinensis extract can better improve the hemorheological parameters and fibrinolytic activities and inhibit the reduction of fibrinolytic activities in the KOA rats.
