CRISPR/Cas9系统联合人多能干细胞制备嵌合体猪培育人源化器官的研究进展

摘要 异源器官移植是解决人类器官资源短缺的一条可行途径,但可供移植的器官明显不足。然而异种器官移植因器官来源广泛,有着非常广阔的前景。猪无论是在基因组水平还是器官形态上,都与人类极为相似,因此一直被认为是最为理想的人类器官的动物供体来源。为了有效的解决人-猪不同种属间的异源器官引发的排斥反应,结合高效特异的CRISPR/Cas9基因组靶向修饰和诱导多能干细胞技术通过囊胚补偿法制备嵌合体猪为培育人类器官成为可能。如若研究证明改方法可行,不仅可有效解决移植因器官源紧缺的问题,同时为研究人类疾病和后续的药物研发提供理想的研究模型。
