摘要
试验旨在从新吉细毛羊皮肤组织表达序列标签(expressed sequence tags,ESTs)中筛选与毛囊发育相关基因并探讨其表达模式与羊毛细度的相关性。利用组装、比对软件Trinity与Blat对新吉细毛羊皮肤组织ESTs组装、注释进而筛选与毛囊发育相关基因,并通过实时荧光定量PCR相对定量方法以2-△△Ct值检测相关基因在不同绵羊个体中的表达规律。结果显示,从474条ESTs中筛选注释获得了4个与毛囊发育相关的角蛋白基因KRT27、KAP3.2、KAP11.1和KAP8.1;4个角蛋白基因在超细型细毛羊皮肤组织表达量极显著高于细型细毛羊的表达量(P<0.01);4个角蛋白基因表达量:KRT27>KAP3.2>KAP11.1>KAP8.1。该研究为角蛋白基因在细毛羊转录水平上的研究提供了一定数据。
The experience was conducted to screen the genes associated with hair follicle development from the expressed sequence tags(ESTs)derived from the skin tissues of Xinji fine wool sheep and explore the association between their expression patterns and the fineness of wool.Using Trinity and Blat,we screened out the candidate genes by assembly and homologous annotation of ESTs,and further studied their expression patterns through detecting the correlation value2^-△△Ct by Real-time PCR method.As a result,four genes(KRT27,KAP3.2,KAP11.1and KAP8.1)were carried out to be responsible for hair follicle development,and 4keratin genes were extremely significant more highly expressed in skin tissues of super fine wool sheep than in fine wool sheep(P〈0.01);The expression level of 4keratin were as follow:KRT27〉KAP3.2〉KAP11.1〉KAP8.1.The study provided certain data for the research on transcript level of fine wool sheep keratin gene.
