摘要 目的 探讨生长发育阶段,早期应激影响C57幼鼠发育的短期实验研究。方法 利用早年生活应激的动物模型干预C57BL/6J母鼠与其幼崽,观察经出生后第2天给予持续早期的慢性应激的C57BL/6J幼鼠,并于第9日龄(P9)、3周龄(P21)时间点检测P 2~9 d幼鼠体质量增加情况;在P 2~9 d期间,每日相同时间段监测1 h内母鼠与幼崽的哺乳情况,幼崽离巢时间及母鼠离巢的次数;在P9、P21日清晨,C57BL/6J幼鼠称质量后颈椎脱臼牺牲幼鼠,分离幼鼠全脑、胸腺及双侧肾上腺并称质量。结果 1经早期慢性应激刺激的C57BL/6J幼鼠,1h观察时间内,每笼至少1只幼崽离巢时间明显高于正常喂养同龄组幼鼠(P〈0.05),相同观察时间内母鼠出巢的次数明显高于对照组(P〈0.05),而1 h观察时间内,母鼠-幼崽的哺乳时间差异无统计学意义(P〉0.1);2经早期慢性应激刺激组幼鼠,在P 2~9 d体质量增加明显低于正常喂养组(P〈0.05),在P9和P21 d,应激组幼鼠的胸腺与体质量比明显低于非应激组(P〈0.05),而双侧肾上腺与体质量比差异无统计学意义(P〉0.05)。结论 发育早期的慢性应激可诱导C57小鼠胸腺过早萎缩。 Objective To explore the short-term experimental study of early stress on C57 pups development in the growth stage. Methods Animal models of early life stress is used to intervene C57BL / 6J dam and pups, and starting from postnatal 2nd day, constant observation C57BL / 6J pups had been subjected to early chronic stress, at P9 and P21 days, measuring pups weight gain from P2 to P9, during P2-9 day. Each day from P2 to P9 days within 1 hour at the same time period, to monitor the time of dam- pups nursing, the number of dams exits and the time of pups outside nest at least one pup. In the morning on P9 and P21 day, after weighing by cervical dislocation handle pups, separating pups global cerebral, thymus and bilateral adrenal and weighed. Results Experienced early chronic stress of C57BL/6J pups, within 1 hour, the number of dam exits and the time of pups outside nest for a pup pups significantly higher than the normal same age group (P〈0.05), but the time of dam-pups nursing showed no statistically significant difference (P〉0.05). Once undergone early chronic stress of pups, the body weight gain is significantly lower than the control group from P2 to P9 (P〈0.05). In the P9 and P21 days, thymus weight compared with the experimental group is significantly lower than the control group (P〈0.05), but bilateral adrenal is no significant difference in body weight. Conclusion Early Chronic stress can induce the C57 mice thymus atrophy prematurely.
