BP1基因对肾透明细胞癌细胞株RLC-310增殖、克隆形成及迁移能力的影响

摘要 目的:研究BP1基因对肾透明细胞癌细胞株RLC-310细胞增殖、克隆形成能力及迁移能力的影响。方法:将前期构建的质粒p EGFP-N1-BP1转染入RLC-310细胞系,用Real-time PCR和Western blot检测BP1 mRNA与蛋白的表达情况;通过MTT、平板克隆实验及Transwell实验检测p EGFP-N1-BP1对RLC-310细胞的作用。结果:转染了p EGFP-N1-BP1的RLC-310细胞中BP1基因/蛋白的表达明显上调;转染组较空白对照组细胞增殖能力明显增加(P<0.05),克隆能力及迁移能力增强(P<0.05)。结论:BP1基因能够增强RLC-310细胞的增殖、克隆形成能力及迁移能力。 Objective :To explore the effects of pEGFP- N1 -BP1 on proliferation, clone formation and migration of RLC -310 cells. Methods:The pEGFP - N1 - BP1 was transfected into RLC - 310 cells. The expression of BP1 gene was detected by Real - time PCR and Western blot. The effects of proliferation, clone formation and migration of RLC -310 cells were examined by MTT, clone formation assay and Transwell assay respectively. Results:The RLC -310 cells transfected with pEGFP - N1 - BP1 plasmid expressed a higher level of Twist protein steadily. The capabili- ties of proliferation, clone formation and migration of RLC -310 cells increased significantly compared with blank con- trol group(P 〈 0.05 ). Conclusion: BP1 gene can enhance the abilities of proliferation, clone formation and migration of RLC - 310 cells.
