摘要
目的:建立拆分阿罗洛尔手性对映体的手性固定相HPLC法。方法:采用纤维素直链淀粉类手性固定相Chiralpak AD-H色谱柱(250mm×4.6mm,5μm),考察了流动相比例、流速、碱性添加剂等因素对阿罗洛尔手性对映体拆分效果的影响。结果:确定了最佳拆分条件:流动相为正己烷-乙醇-二乙胺-三乙胺=30∶70∶0.1∶0.1(V/V),流速为0.4ml/min,检测波长为315nm,柱温为25℃,阿罗洛尔对映体拆分的分离度为3.033,对映体能够得到良好的拆分。结论:该法操作简便,可用于阿罗洛尔对映体的分离及质量控制。
Objective: To establish a chiral stationary phase HPLC method for the enantiomeric separation of arotinolol. Methods: An amylose chiral stationary phase Chiralpak AD-H column (250 min× 4.6 mm,5 μm) was used. Effects of the pro- portion of the mobile phase, flow rate and alkaline additives on the separation of arotinolol enantiomers were investigated. Results:The optimized chromatographic conditions were as follows: the mobile phase was hexane : ethanol : diethylamine : triethylamine:30 : 70 : 0. ] : 0. I(V/V) with a flow rate of 0.4 ml/min and the detection wavelength of 315 nm. The column temperature was set at 25 ~C. The arotinolol enantiomers could be well separated and the resolution was 3. 033. Conclusion: This method is simple to handle and it could be used for the separation and quality control of arotinolol enantiomers.
