摘要
目的观察复肾汤对db/db小鼠肾脏病变的影响。方法正常对照组为雄性C57小鼠8只,常规饲养。雄性db/db鼠24只,正常饲养1周,血糖稳定升高4周后分为模型组、复肾汤小剂量组(L1)、复肾汤大剂量组(L2)。模型组灌胃给予生理盐水;L1组按FST 13mg/(kg·d)灌胃;L2组按FST 39mg/(kg·d)灌胃;连续8周,定期测体质量、血糖。实验结束后,采集血清,检测T-chol、BUN、SOD、MDA等生化指标;轻取肾脏,免疫组织化学法检测肾组织VEGF、FN的表达,光镜下观察肾脏组织病理变化。结果与模型组比较,L1、L2组的T-chol、BUN、MDA明显降低(P〈0.05-0.01),SOD明显升高(P〈0.05),VEGF、FN表达明显下调,病理学评分明显降低(P〈0.05)。结论复肾汤可以明显改善db/db小鼠肾脏损害,具有延缓糖尿病肾病病情发展的作用。
OBJECTIVE To observe the effects of Fushen Decotion on renal lesion in db/db mice.METHODS 8male C57 mice were chosen as normal control group,conventional breeding.24 male db/db mice with high blood glucose level were divided into model group,FST(L1)group and FST(L2)group.Model group mice were given normal saline;L1and L2 mice were treated with FST 13g/kg and 39g/kg,respectively.The weight and blood sugar of mice were measured regularly for 8weeks.After the experiment,the serum was collected and biochemical parameters,such as T-chol,BUN,SOD and MDA,were detected.Then the kidneys were taken,the expression of VEGF and FN in renal tissues were detected by immunohistochemical method,and the renal lesions were observed under light microscope.RESULTS Compared with the model group,T-chol,BUN and MDA of L1 and L2groups were significantly decreased(P〈0.05-0.01),SOD was increased significantly(P〈0.05),expression of VEGF and FN decreased significantly,and pathology score decreased significantly(P〈0.05).CONCLUSION Fushen Decotion can significantly improve the db/db mice renal damage and delay the development of diabetic nephropathy.
