哈瓦那辣椒悬浮培养的体细胞胚发生
摘要
本文对不同培养基、不同外植体类型(节点、节间、下胚轴、半粒种子、果段)诱导哈瓦那辣椒产生体细胞胚胎的情况进行了评估。在添加9.05μM 2,4-D、3%蔗糖和0.8%植物凝胶的MS基本培养基中,25±2℃,连续30d光照(40~50μmol·m^2/s)条件下,诱导胚性愈伤组织。形成的愈伤组织将被转移到组成相同的液体培养基中。仅当以下胚轴外植体,用含3.4IxM噻苯隆的MS培养基对其进行诱导时,才能产生体细胞胚。本研究建立了辣椒“细胞悬浮培养的体细胞胚发生”的一种实验方案。
