Establishment of Rat Model with Chronic Obstructive Pulmonary Disease of Yindeficiency and Phlegm-retention Syndrome Type

摘要 目的:建立阴虚痰饮证慢性阻塞性肺疾病大鼠模型。方法:利用烟熏、气管内滴注脂多糖、地塞米松磷酸钠注射液肌注的方法制造慢性阻塞性肺疾病阴虚痰饮大鼠模型,并用金水六君煎干预进行反证,观察大鼠一般情况、肺组织病理学改变、血气分析、血清肿瘤坏死因子、血清皮质酮、促肾上腺皮质激素水平。结果:慢性阻塞性肺疾病阴虚痰饮证大鼠一般情况与临床阴虚痰饮证表现相符,血清皮质酮、促肾上腺皮质激素水平高于慢性阻塞性肺疾病大鼠模型,氧分压低于慢性阻塞性肺疾病大鼠,二氧化碳分压高于慢性阻塞性肺疾病大鼠;金水六君煎干预后症状好转,血气分析结果改善。结论:用本实验方法建立慢性阻塞性肺疾病阴虚痰饮证大鼠模型是可行的。 Objective: To study the method of establishing rat model with chronic: obstructive pulmonary di'sease (GOPD) ofVin-deficiency and phlegm-retention syndrome type. Methods: The rat models were established by fumigation combined with en- dotracheal instillation of lipopolysaccharide and intramuscular injection of dexamethasone sodium phosphate inj6cti0n. Jinshui Ll-ujun Decoction(金水六君煎) was used as intervention factor to the rat models. General conditions and pathological changes of lung tissue were observed, and blood gas analysis, the levels of serum tumor necrosis factor, corticosterone and adrenocorti- cotropic hormone were detected. Results: The general conditions of rat models were consonant with clinical manifestation of Yin-deficiency and phlegm-retention syndrome type. The contents of corticosterone and adrenocorticotropic hormone in COPD rat models with Yin-deficiency and phlegm retention syndrome were higher than those in COPD rat models, with higher oxygen partial pressure and lower carbon dioxide partial pressure, which were improved by the invention of Jinshui Liujun Decoction. Conclusion: The method is feasible in replicating COPD rat model with Yin-deficiency and phlegm retention syndrome.
