不同产地、不同品种间绞股蓝总皂苷和总多糖含量比较研究 被引量:7

摘要 目的:建立绞股蓝中总皂苷、总多糖含量测定方法,比较不同产地、不同品种绞股蓝中总皂苷、总多糖含量差异,为考察药材内在质量提供依据。方法:采用紫外-可见分光光度法,以人参皂苷Re为对照品,于550nm波长处对绞股蓝总皂苷吸光度进行测定;以D-无水葡萄糖为对照品,于490nm波长处对绞股蓝总多糖吸光度进行测定。结果:人参皂苷Re在0.097 8--0.489mg范围内与其吸光度呈良好的线性关系(R2=0.999 8),平均回收率为97.79%,RSD为1.44%(n=6);D-无水葡萄糖在0.0198~0.099mg范围内与其吸光度呈良好的线性关系(R2=0.999 4),平均回收率为99.58%,RSD为1.35%(n=6)。结论:该方法简便可行、重复性好,适用于绞股蓝中总皂苷、总多糖含量测定。不同产地绞股蓝中总皂苷和总多糖含量差异较大,且不同品种间绞股蓝总皂苷含量存在显著性差异,但绞股蓝总多糖含量与绞股蓝品种无相关性。该结果可为绞股蓝的质量控制和相关产品的开发研究提供参考依据。 Objective:Take the content of total saponins and polysaccharides as index, to establish a method for assaying the sapo- nins and polysaccharides, to compare the diversity of total saponins and polysaccharides content inGynostemma pentaphyllurn (Thunb.) Makino from different producing areas and different varieties, to provide basis for the study of its internal quality. Meth- ods: The UV spectrophotometry was used to determine the contents of total saponins and polysaccharides. The detection wavelength are 550nm and 490nm respectively. The reference substance are Ginsenosides Re and D-Glucose. Results. Good linear relationship with absorbability occurred when Ginsenosides Re in the range of 0. 097 8--0. 498mg (R2 =0. 999 8), and the average recovery was 97.79% (RSD=1. 44%, n=6). Good linear relationship with absorbability occurred when D-Glucose in the range of 0. 019 8- 0. 099mg (R2 =0. 999 4), and the average recovery was 99.58% (RSD= 1.35%, n=6). Conclusion: The established method is simple, accurate and can be used for the determination of total saponins and polysaccharides. The total saponins content exist differ- ences inGynostemma pentaphyllum (Thunb.) Makino from different producing areas and different varieties. The total polysaccharides content exist differences in Gynostemma pentaphyllum (Thunb.) Makino from different producing areas, but the content of total polysaccharides have no correlation with varieties. The results can able to provide reference for quality control of Gynostemma pentaphyllum (Thunb.) Makino and related product development research.
