摘要
采用优化的实验条件建立高效液相色谱法,测定荞麦种质中的芦丁含量。称取一定量干燥后的荞麦样品粉末,溶于甲醇,于60℃水浴中超声提取2 h,将提取液定容后,上高效液相色谱仪分析检测。色谱条件为:安捷伦C18柱(4.6 mm×150 mm,5μm),柱温30℃,流动相甲醇∶0.5%磷酸(V/V)为40∶60,流速1 m L/min,进样量10μL,检测波长257 nm。结果表明,该方法准确、灵敏、重现性好,为芦丁提取及进一步鉴选高芦丁含量的荞麦种质奠定了基础。
Using the optimized conditions, the determination of buckwheat rutin was set up by high performance liquid chromatog- raphy. Buckwheat sample was weighed amount of powder after drying, the addition of methanol, in ultrasonic extraction at 60 % in 2 h, the extracted fluid volume, on HPLC analysis and detection. Chromatographic conditions was as follows: Agilent C18 column (4.6 mm × 150 mm, 5μm), column temperature was 30 ℃, the mobile phase methanol : 0.5% phosphoric acid( V/V ) was 40 : 60, flow rate was 1 mL/min, the injection volume was 10 μL, the detection wavelength was 257 nm. The results showed that the method was accurate, sen- sitive, reproducible, but also for the identification and selection of rutin extraction and further high rutin buckwheat laid the foundation.
