糖尿病心肌病大鼠心肌中偶联因子6的变化及意义

Changes and significance of coupling factor 6 in the myocardium of diabetic cardiomyopathy rats

摘要 目的探讨偶联因子6(CF6)在糖尿病心肌病大鼠心肌中的变化及其意义。方法采用链脲佐菌素(STZ)建立糖尿病心肌病大鼠模型,同时采用正常大鼠作为对照组,检测并比较两组大鼠的心率、收缩压以及生化指标,测量并计算两组大鼠的左心系数,Masson三色染色观察心肌组织胶原含量变化,评估心肌重构情况,进一步应用Western blot技术检测并比较两组大鼠心肌组织中CF6的表达情况。结果模型组心率(HR)及收缩压(SBP)与对照组比较差异均无统计学意义(P>0.05);模型组体质量、空腹血糖(FBG)均高于对照组,差异有统计学意义(P<0.01和P<0.05)。模型组大鼠的左心系数明显高于对照组,差异有统计学意义(P<0.01),模型组大鼠心肌组织中胶原纤维排列紊乱粗大,含量增加,模型组大鼠心肌中CF6的表达明显高于对照组,差异有统计学意义(P<0.05)。结论 CF6在糖尿病心肌病大鼠心肌组织的大量表达,提示其可能参与心肌重构起并起重要作用。 Objective To study the coupling factor 6 (CF6) in diabetic cardiomyopathy in rats and its significance in the myocardium. Methods Chain urea with cephalosporins (STZ) diabetic eardiomyopathy in rats model was established, at the same time using normal rats as control group, test and compare the heart rate and systolic blood pressure and biochemical indexes of two groups, the results of Masson staining and LVW/BW (left ventricular weight/body weight) calculation were evaluated to observe the pathological change and myocardial remodeling of myocardium tissue. Further applied Western blot technique to detect and compare myocardial tissue CF6 expression of the two groups. Results The heart rate (HR) and the systolic blood pressure (SBP) of the model group compared with control group had no statistical significance (P〉0.05); body quality and fasting blood glucose (FBG) of model group were higher than the control group, the difference was statistically significant (P〈0.01 and P〈0.05). The cardiomyocytes of rats in model group ranged disorderly and loosely, and the collagenous fiber in rats increased obviously. Rats left heart coefficient of the model group was obviously higher than that of control group, the difference was statistically significant (P〈0.01), in the model group rats myocardial CF6 expression was significantly higher than the control group, the difference was statistically significant (P〈0.05). Conclusion The high CF6 expression of myocardial tissue in diabetic cardiomyopathy rats prompt that it may participate in myocardial remodeling and play an important role.
