毛木耳子实体发育相关cDNA差异显示分析

摘要 以毛木耳菌丝、耳基和耳片为供试材料,采用3’-c DNA限制性片段展示方法,获得3个阶段c DNA表达谱,对大于100 bp的18个c DNA片段使用blastx程序与非冗余蛋白数据库(non-redundant protein sequence)进行同源性比对。结果表明,有6个序列能够分别与NCBI数据库中的新孢子虫假定蛋白、水霉菌Aphanomyces invadans蛋白TAR1、微绿球藻核糖体RNA反义转录序列、双孢蘑菇、糙皮侧耳、皱木耳假定蛋白匹配蛋白质相匹配,并且E值≤5×10-8,表明序列匹配良好,实验方法可行,能够作为毛木耳等食用菌功能基因研究手段之一。 Hypha,primordium and fruiting body of Auricularia nigricans were used to study their gene expression by display of differential developmental 3'end-c DNAs. And 18 c DNA fragments( ≥100 bp) were translated and used to align non-redundant protein sequences in NCBI with blastx. Results showed that six sequences were significantly aligned to the proteins of Nannochloropsis gaditana transcript antisense to ribosomal rna protein,Agaricus bisporus hypothetical protein AGABI1DRAFT_49393,Pleurotus ostreatus hypothetical protein PLEOSDRAFT_23300,Auricularia delicata hypothetical protein AURDEDRAFT_115833,Aphanomyces invadans protein TAR1,Neospora caninum hypothetical protein NCLIV_068840,and all E values ≤5 × 10- 8. This indicated that the experimental method was feasible and could be used as a tool for the function gene study of mushroom such as Auricularia nigricans.
