摘要
microRNAs(miRNAs)在调节动物的获得性和先天性免疫中发挥重要作用。为研究病毒感染对凡纳滨对虾miRNA表达的影响,构建了桃拉综合症病毒感染和对照的凡纳滨对虾的小RNA文库,利用Solexa高通量测序技术对小RNA文库进行测序,分别得到61854276和65320117条小RNA序列,其中有2864337条序列被注释为保守的miRNA,对应87种已知的miRNA。对高表达量的miRNA进行表达差异分析,得到了22个差异表达miRNA,其中8个上调表达,14个下调表达。对差异表达miRNA的靶基因进行了预测,其中预测得分最高的靶基因大多数涉及到动物的免疫或细胞凋亡功能,说明miRNA在对虾的免疫机制中起着非常重要的作用。研究结果提供了凡纳滨对虾miRNA响应病毒感染的表达谱的基础资料,这将有助于揭示miRNA在对虾的抗病毒免疫机制中的作用。
microRNAs( miRNA) play an important role in regulating both adaptive and innate immunity. Litopenaeus vannamei is the world's most widely farmed crustaceans. To study the effect of viral infection on miRNA expression in Litopenaeus vannamei,we constructed two small RNA libraries from Taura syndrome virus infected shrimp and control shrimp,respectively,and used Solexa high-throughput sequencing technology to sequence the small RNA libraries. In total,61,854,276 and 65,320,117 small RNA sequences were obtained,respectively,in which 2,864,337 sequences were annotated as conserved miRNA,corresponding to 87 kinds of known miRNAs. The differential expression of the highly expressed miRNAs were analysed,and 22 differentially expressed miRNAs were obtained,in which 8 were upregulated and 14 were downregulated. The target genes of the differentially expressed miRNAs were predicted,and the majority of the highest scoring target genes were involved in the immune or apoptosis function of animals,indicating that miRNAs play a very important role in the immune system of shrimp. This study provides basic information of miRNA expression profiles of Litopenaeus vannamei in response to viral infection,which will help to reveal the role of miRNA in antiviral immune mechanism in shrimp.
