摘要
目的建立HPLC同时测定氯霉素氢化可的松滴耳液中氯霉素和氢化可的松含量的方法。方法色谱柱:Wonda Sil(C18-WR;4.6 mm×250 mm;5μm);流动相:乙腈-0.1%庚烷磺酸钠溶液;流速:1.0 ml/min;检测波长:0~16 min为278 nm,16~20 min为245 nm,20~25 min为278 nm;柱温:30℃;进样量:20μl。结果氯霉素线性回归方程为Y=27.021X+9.71,r=1(n=6);氢化可的松线性回归方程为Y=40.867X-7.53,r=0.9999(n=6);氯霉素平均回收率为99.1%,RSD=0.4%;氢化可的松平均回收率为101.5%,RSD=1.1%。结论 HPLC法操作方便,定量准确,可用于同时测定该制剂中两组分的含量。
Objective To establish an HPLC method for simultaneous determination of content of chloramphenicol and hydroeortisone in compound chloramphenicol hydrocortisone auristillae. Methods Chromatographic column was WondaSil C18 column (C18-WR;4.6 mm×250 mm;5 μm); ,nobile phase consisted of acetonitrile-0.1% heptane sodium sulfonate solution; the flow rate was 1.0 mi/min, the detection wavelength was 278 nm at 0-16 min, 245 nm during 16-20 min, and finally 278 nm in the rest 20-25 min; the column temperature was 30 ℃; the injection volume was 20 jxl. ll.esults The calculated linear regression equation of ehloramphenicol was 1=27.021X+9.71, r=1 (n=6); that of hydrocortisone was Y=40.867 X-7.53, t=0.9999 (n=6). The average recovery rates of chloramphenieol and hydroeortisone were 99.1% (RSD=0.4%) and 101.5% (RSD=1.1%), respectively. Conclusion The HPLC method is simple and accurate, which can be used in the simuhaneous determination of content of ehloramphenicol andhydroeortisone in this preparation.
