大黄中蒽醌类化合物对大鼠肠平滑肌条收缩作用影响的研究 被引量:6

摘要 目的:比较大黄中五种主要的蒽醌类化合物对大鼠游离空肠平滑肌条(JSMF)和结肠平滑肌条(CSMF)收缩作用的影响。方法:取正常SD大鼠制备游离JSMF及CSMF,观察大黄素、大黄酸、大黄酚、大黄素甲醚和芦荟大黄素对其收缩作用及其结构对药效的影响。结果:大黄素对JSMF及CSMF的收缩有双向调节作用;大黄素甲醚和大黄酸可抑制JSMF收缩,并呈剂量依赖性;5种蒽醌化合物在浓度超过20μmol/L时,均可抑制CSMF收缩。结论:5种蒽醌化合物对肠道平滑肌条收缩的影响可能与其7位羟基和3位羧基及取代基的极性、立体结构、受体的分布有关。 Objective: To investigate effects of Anthraquinone compounds from rhubarb on contractility of isolated jejunal smooth muscle fragment( JSMF) and colon smooth muscle fragment( CSMF) in rats. Methods: Jejunum and colon were isolated from normal SD rats and JSMF and CSMF were made. The effects of emodin,rhein,chrysophanol,physcion and aloe-emodin on them were observed and analyzed. Results: The results showed emodin had a dual-directional regulation on the contractility of JSMF and CSMF;physcion and rhein inhibited the contractility of JSMF in a dose dependent manner; and these Anthraquinone compounds showed inhibitory effects on the contractility of CSMF over 20μM. Conclusions: The effects of these Anthraquinone compounds on the contractility of JSMF and CSMF may be related to hydroxyl radical at 7 position and carboxyl group at 3 position,polarity and special stereochemical structure of substituent groups,and distribution of receptors in jejunum and colon.
