摘要 多重链接探针扩增技术是将核酸杂交和PCR链式扩增相结合的一项高通量检测技术。该技术特异性强、敏感性高,可以实现多达50个靶分子的同步检测。自2002年被报道以来,该技术取得了很大进展,目前已广泛用于遗传性疾病的基因检测、肿瘤预后及传染病高通量检测领域。本文就多重链接探针扩增技术的原理、优缺点及应用等进行了探讨。 Multiplex ligation-dependent probe amplification(MLPA)is a high throughput detection technology combining nucleic acid hybridization with PCR chain amplification. With it's high specificity and high sensibility,MLPA could achieved synchronous detection of as many as 50 target molecules. Since 2002,the technology has made a great progress,and been extensively used in gene detection of genetic disease and tumor prognosis,and high-throughput detection of infectious diseases. The principle,advantages and disadvantages,application of MLPA were discussed in this paper.
