右美托咪定对喹啉酸引发的成年大鼠空间记忆损害的影响作用

The Effect of Dexmedetomidine on Impairment of Spatial Memory of Adult Rats Induced by Quinolinic Acid

摘要 目的探讨右美托咪定对喹啉酸引发的成年大鼠空间记忆损害的影响作用及其可能机制。方法选择健康成年Wistar雄性大鼠30只,随机数字法分为生理盐水组(NS组)、喹啉酸组(QA组)、右美托咪定+喹啉酸组(D+Q组)。八臂迷宫实验、旷场实验方法测定各组大鼠的空间记忆能力、探究行为的变化。实验完成后立即处死大鼠,提取海马组织,应用Western Blotting法测定海马内胆碱乙酰转移酶(choline acetyltransferase,ChAT)蛋白的表达水平。结果与NS组比较,QA组、D+Q组的空间记忆功能明显减退,自发性活动减少,ChAT蛋白表达明显下降(P<0.05)。与QA组比较,D+Q组的空间记忆功能有所改善(P<0.05)。但自发性活动无明显改变(P>0.05),ChAT表达有所增加(P<0.05)。结论 QA所致空间记忆障碍可能与海马内ChAT表达下调有关系,右美托咪定可部分逆转ChAT表达下调。 Objective To evaluate the effect of dexmedetomidine on impairment of spatial memory of adult rats induced by quin -olinic acid and its possible mechanism .Methods Thirty healthy adult male Wistar rats were randomly divided into 3 groups:normal saline (NS group),quinoline acid group(QA group) and dexmedetomidine plus quinoline acid group (D+Q group).Eight-arm radial maze task and open-field test was detected to evaluate the spatial memory and exploratory behavior .Once the test were finished ,rats were killed to meas-ure the expression of ChAT of hippocampus by Western Blotting methods .Results Compared with NS group ,spatial memory and spontaneous activity in QA group and D +Q group was declined significantly,the expression of ChAT decreased (P〈0.05).Compared with QA group, spatial memory in D+Q group was improved significantly (P〈0.05).Spontaneous activity did not change significantly (P〉0.05).The ex-pression of ChAT increased significantly (P〈0.05).Conclusion Spatial memory impairment induced by QA may be associated with the low expression of ChAT of hippocampus .Dexmedetomidine may partially reversed the low expression of ChAT .
