摘要
目的观察骨髓间充质干细胞(bone marrow mesenchymal stem cells,BMMSCs)移植后对急性胰腺炎大鼠胰腺损伤的修复作用,并探讨其治疗机制。方法体外培养、扩增BMMSCs并用荧光染料PKH26进行标记。采用L-精氨酸诱导急性胰腺炎模型,实验随机分为对照组和干细胞移植治疗组(BMMSCs移植治疗组)。移植后,被PKH26标记的BMMSCs出现在损伤的胰腺组织中,通过免疫荧光和免疫组化检测胰腺组织表面标记物Pax-4、Ngn3来判定BMMSCs的定植。结果病理组织学检查证实L-精氨酸诱导大鼠急性胰腺炎模型成功;移植术后3周,在大鼠急性胰腺炎损伤胰腺中可检测出PKH26标记的荧光细胞,PKH26标记的细胞表达胰腺组织表面标志物包括Pax-4、Ngn3。结论 BMMSCs移植可以促进损伤胰腺组织的再生。
Objective To observe the recovery effect and treatment mechanism of bone marrow mesenchymal stem cells (BMMSCs) transplantation in the pancreas injury of acute pancreatitis of rats. Methods BMMSCs were isolated, cultured and identified in vitro and labeled with the fluorescent dye PKH26. Acute pancreatitis rat model was induced by L-arginine. The experiment was divided into control group and BMMSCs treatment group. Transplanted PKH26 positive BMMSCs were presented in the injured pancreas, and the markers of pancreas including Pax-4, Ngn3 were detected by immunofluorescence and immunohistochemical. Results Histopathological examination confirmed that L-arginine in- duced acute pancreatitis model of the rat was successful. PKH26 fluorescent cells were detected about 3 weeks post- transplantation in the acute pancreatitis model rats. PKH26 labele cells expressing pancreas cells markers included Pax- 4, Ngn3. Conclusion The BMMSCs transplantation could promote self-restoration and pathological regeneration in pancreatic injury.
