舒郁散对慢性应激抑郁大鼠行为及脑PSD-95蛋白表达影响

The Effect of Shuyusan Scatter on Concentrations of Brain Cortex Neurotransmitters and PSD-95 Expression with Chronic Mild Unpredictable Stressors Depression Rats

摘要 目的:观察舒郁散对慢性不可预见性应激(CUMS)抑郁大鼠行为、大脑皮层去甲肾上腺素(NE)、多巴胺(DA)、5-羟色胺(5-HT)及突触后致密物95(PSD-95)蛋白表达的影响。方法:50只大鼠随机分为:正常对照组、CUMS模型组、模型加盐酸氟西汀片治疗组、模型加舒郁散小剂量、大剂量治疗组。对大鼠进行21 d的慢性不可预见性应激刺激建立模型,采用糖水实验、悬尾实验观察大鼠行为改变,用ELISA法检测大鼠大脑皮质神经递质的含量,用免疫组织化学染色法观察大鼠大脑皮质PSD-95蛋白表达。结果:盐酸氟西汀组、舒郁散大剂量组大鼠悬尾不动时间明显延长,糖水消耗百分率明显下降,与模型组比差异显著,P<0.01。盐酸氟西汀、舒郁散大剂量组大鼠尾悬不动时间明显减少,糖水消耗量明显增加,与模型组比差异显著,P<0.05。模型组鼠脑组织NE、5-HT的含量显著降低,与正常对照组比差异显著,P<0.05;大剂量舒郁散能显著提高CUMS大鼠脑内NE、5-HT的含量,与模型组比差异显著,P<0.05。模型组大鼠大脑皮层PSD-95蛋白表达减少,与正常组比差异显著,P<0.05;大剂量舒郁散能显著提高CUMS大鼠脑内PSD-95蛋白表达,与模型组比差异显著,P<0.05。结论:动物行为学显示舒郁散具有抗抑郁作用,这种作用与提高慢性应激抑郁大鼠脑内神经递质的含量、提高大脑皮层PSD-95表达有关,上述作用与其剂量呈正相关。 Objective:To investigate the effect of Shuyusan Scatter on the animal behavior,as well as the level changes of norepinephrine(NE),dopamine(DA),serotonin(5-HT)and postsynaptic density protein 95(PSD-95)expression in brain cortex with chronic mild unpredictable stress depression(CMUS)rats. Method:50 mice were randomly divided into 5 groups:normal control group,CMUS model group,fluoxetine group,and high/Low dose group of Suyusan. Except normal group,the other groups were given CMUS for 21 days. Tail suspend test(TST)and sucrose preference test(SPT)were used to estimate animals' behavior. The content level of brain cortex neurotransmitter was determined by enzyme linked immunosorbent assay(ELISA). PSD-95 protein expression in brain cortex was detected. Results:The fluoxetine treatment group and high dose of Shuyusan Scatter group showed significant increase in the SPT and decrease in the immobility time in TST,there was significant difference compare with CMUS model group,P〈0.05. The levels of NE and 5-HT were significant reduced in model group,compare with normal group,P〈0.05. High dosage Shuyusan Scatter group and fluoxetine could significantly increase NE and DA level,compared with the model group P〈0.05. The positive cell number of PSD-95 expression in brain cortex was significantly decreased in model group,compared with normal group P〈0.05. High dosage Suyusan could significantly increase PSD-95 expression,compared with the model group P〈0.05. Conclusion:Suyusan Scatter has anti-depression effect,this effect connected with increase NE and 5-HT contents of mice cerebrum in cortex and increases PSD-95 expression,the effect is increased as concentration increased.
