参麦注射液对犬冠状动脉环舒缩功能影响

摘要 目的:观察参麦注射液对犬冠状动脉环舒缩作用的影响并探讨其作用机制。方法:6只犬麻醉处死后取冠状动脉并制成2 mm宽的动脉环,将动脉环标本随机分为对照组、参麦组,以去甲肾上腺素(NE)和氯化钾(KCl)分别灌流两组犬离体冠状动脉环,预激使之达到最大收缩峰值,再分别加入10-5~10-1 mol/L不同浓度的参麦注射液,测定参麦注射液对犬冠状动脉环收缩及舒张张力,并计算其半抑制浓度(IC50)。结果:10-3~10-1 mol/L参麦注射液能明显抑制NE和KCl引起的离体冠脉收缩,降低收缩及舒张张力。与药前及对照组比较,P〈0.05,差异均有统计学意义。结论:参麦注射液可抑制NE和KCl引起的冠脉收缩,具有α受体阻断效应,其作用机制可能是抑制犬冠状动脉平滑肌细胞上分布的受体操控钙通道及电压依赖性钙通道有关,有抑制平滑肌细胞的内钙释放和外钙内流的作用。 Objective:To observe the effect of Shenmai Injection on canine coronary artery rings of vasomotor function and to explore its mechanism of action. Methods:6 dogs were sacrificed after coronary artery and arterial ring made of 2 mm wide,the arterial rings were randomly divided into control group,Shenmai group,in order to norepinephrine(NE)and potassium chloride(KCl)were perfused two groups of isolated canine coronary artery rings,pre excitation to achieve maximum contraction peak value,and then to10-5-10-2 mol/L Shenmai Injection perfusion,determination of Shenmai injection in canine coronary vasomotor tone ring and calculate the half inhibitory concentration(IC50). Results:Isolated coronary artery constriction of 10-3-10-1 mol/L Shenmai inhibited NE and KCl induced contraction,reduce tension. And before treatment and the control group,P〈0.05,there was significant difference. Conclusion:Coronary artery constriction and michael inhibited NE induced and KCl,which may release and calcium current and the inhibition of smooth muscle cell voltage dependent calcium channels and gymnastics longitudinal calcium channel mediated calcium. Inhibit smooth muscle cell intracellular calcium release and calcium influx role.
