摘要
建立赤狐的耳成纤维细胞的分离和培养体系,为狐的核移植,转基因狐的研究奠定基础。采用组织块培养法分离狐耳成纤维细胞的原代细胞,再利用胰酶短暂消化法进行纯化传代培养,观察成纤维细胞的形态,并进行染色体分析。结果表明,组织块培养法可获得成纤维细胞,经过纯化培养,可获得均一、稳定的狐耳成纤维细胞。可以作为核移植的供体细胞,推动特种动物的转基因研究。
This study is to isolate and culture skin fibroblasts from the ear of fox. It propels the research on fox somatic nuclear transfer and transgenic fox. In this study,fox ear skin fibroblasts were obtained by tissue pieced culture. The primary culture was digested by trypsin at a short time for the subculture. The fibroblasts were observed by the electron microscope,which had a good growth status. Then karyotype analysis was conducted on the fibroblasts,which showed a standard karyotype of fox. The results showed that fox ear skin fibroblasts could be cultivated successfully in vitro by means of tissues culture method. The primary culture could be purified and ear fibroblast cell line be formed,which can be used as donor cell for nuclear transfer and propels the research on transgenetic special animals.
