菩人丹对糖尿病大鼠视网膜HO-1表达的作用 被引量:1

摘要 目的:探讨菩人丹(PRD)对糖尿病大鼠视网膜血红素加氧酶l(HO-1)表达的影响。方法:36只雄性Wistar大鼠随机分为正常对照组、糖尿病模型组和PRD治疗组,每组12只。糖尿病模型组、PRD治疗组大鼠均采用链脲佐菌素连续腹腔注射的方法建立2型糖尿病大鼠模型,模型成功建立后,PRD治疗组大鼠给予PRD(1.8g/k g/d)灌胃3个月。分别采用Western blot法和RT-PCR法检测大鼠视网膜HO-1蛋白和m RNA的表达。结果:模型组大鼠视网膜HO-1蛋白和m RNA的表达明显高于正常对照组大鼠(P<0.01),PRD治疗组大鼠视网膜HO-1蛋白和m RNA的表达明显高于模型组大鼠(P<0.01)。结论:PRD可通过上调HO-1的表达减轻糖尿病时视网膜氧化应激损伤,发挥对糖尿病视网膜病变的保护作用。 Objective: To investigate the effects of Purendan(PRD) on heme oxygenase-1(HO-1) expression in retina of diabetic rats. Methods: 36 male Wistar rats were randomly divided into 3 groups(n=12): normal control group,diabetes model group,PRD treatment group. The diabetes model rats were made by continuous intraperitoneal injection of streptozotocin. After the diabetes animal model was successfully established, the rats in PRD treatment group were lavaged with PRD(1.8g / k g / d) for 3 months. Western blot and RT-PCR were respectively used to detect the expression of HO-1 protein and m RNA in retina of rats. Results: The expression of HO-1 protein and m RNA in retina of rats in diabetes model group were obviously higher than normal control rats(P〈0.01). Compared with diabetes model rats, the expression of HO-1 protein and m RNA in retina of rats in PRD treatment group increased signifi cantly(P〈0.01). Conclusions: PRD can reduce oxidative stress injury in retina by up-regulating HO-1 expression in retina, thus has protective effects on diabetic retinopathy.
