摘要
研究了裂蹄针层孔菌(Phellinus linteus)发酵菌丝体对人肝癌Hep G-2细胞的抑制作用。采用MTT比色法测定不同浓度发酵菌丝体对肝癌细胞存活率的影响;建立荷瘤Hep G-2裸鼠模型,检测不同剂量裂蹄针层孔菌发酵菌丝体对肿瘤的抑制作用。MTT法观察结果显示,裂蹄针层孔菌发酵菌丝体可抑制人肝癌Hep G-2细胞的增殖,且呈现剂量效应关系,高剂量(960 mg·L^(-1))能显著抑制肿瘤细胞的增殖,最佳抑制时间为24 h;裂蹄针层孔菌发酵菌丝体中剂量(500 mg/kg·d)、高剂量(1 000 mg/kg·d)对荷瘤Hep G-2裸鼠有显著的抑瘤作用,抑瘤率分别为40.42%、35.06%。裂蹄针层孔菌发酵菌丝体对人肝癌Hep G-2肿瘤细胞有较好的抑制作用。
The inhibition effect on HepG-2 of Phellinus linteus mycelium was studied. The survival rates of cancer cell by dif- ferent concentration myeelium using MTT method were determined, and through establish tumor bearing HepG-2 mice model, the inhibition effect of different dose mycelium on tumor was studied. The results demonstrated that mycelium could inhibit HepG-2 cell proliferation in a dose-dependently, high dose of 960 mg· L^-1 could significantly suppress cancer cell proliferation with the optimum time of 24 hours. The mycelium also have outstanding tumor suppression on tumor bearing HepG-2 mice, with inhibitory rates of 40.42% and 35.06% at the respective doses of 500 mg/kg·d and 1000 mg/kg·d. The fermentation mycelium of P. linteus had stronger inhibition effect on HepG-2.
