摘要
为从自然界寻找高效的红枣果胶酶生产菌,以红枣果胶为唯一碳源,从保定市阜平县采集的腐烂红枣、土壤等中筛选、分离纯化出一株高产果胶酶的菌株,编号为M3,该菌株在30℃摇床转速160 r/min条件下培养72 h,其果胶酶活性可达188.89 U/m L;采用CTAB法提取其基因组总DNA,利用真菌的通用引物ITS1和ITS4扩增菌株的r DNA ITS区序列,扩增产物纯化后进行测序。将测序结果在Gen Bank中进行同源性搜索,并下载部分具有代表性菌种的ITS序列,利用软件MEGA6构建系统发育树,通过序列分析,该菌株鉴定为白地霉(Galactomyces candidum)。该菌株的ITS序列基因已登录Gen Bank,登录号为:KP399955。
The aim of this work was to screen highly jujube pectinase producing bacteria for following laboratorial research and industrial application. A high-yield pectinase strain M3 was screened from Rotten jujube, soil, etc from Fuping county of Baoding city. This strain was cultured at the condition of 30 ℃, 160 r/ min for 72 h, its pectinase activity can reached to 188.89 U/mL. Morphological and molecular identification of M3 was carried out by CTAB method to extract its genomic DNA, use fungal universal primers of ITS1 and ITS4 to amplify the strain's rDNA ITS sequences. The amplified product was sequenced after purification. Carry out the results by homology searching in GenBank. Download some representative species of ITS sequences. Using software of MEGA6 build molecular phylogenetic tree, the strain was identified as Galactomyces candidum. The ITS sequence of the bacterium gene is logged in GenBank, registreation number was KP399955.
