摘要
目的:探讨脑通汤对缺氧-复氧大鼠海马神经元脑源性神经营养因子基因(BDNF mRNA)表达的影响。方法:将体外培养大鼠海马神经元分为正常细胞组、缺氧-复氧模型组、正常血清组和脑通汤大、中、小剂量含药血清组,实时荧光定量PCR法检测脑通汤干预缺氧24 h复氧2 h后海马神经元BDNF mRNA的表达。结果:实时荧光定量PCR结果显示,与正常海马细胞组比较,缺氧-复氧模型组海马BDNF mRNA的相对表达量明显下降(P<0.05);与缺氧-复氧模型组海马细胞相比,脑通汤大、中、小剂量含药血清组海马细胞中BDNF mRNA的相对表达量明显增高(P<0.05),其中脑通汤大、中剂量含药血清组升高更明显(P<0.05);但正常血清组与模型组海马细胞比较BDNF mRNA的表达趋势差异无统计学意义(P>0.05)。结论:脑通汤可以上调缺氧-复氧后海马神经元BDNF mRNA的表达。
Objective: To investigate the effect of Naotong soup on the expression of brain-derived neurotrophic factor( BDNF) mRNA in hypoxia-reoxygenation rat hippocampal neurons. Methods:Hippocampal neurons of rats were cultured in vitro,then divided into normal cell group,hypoxia-reoxygenation model group,normal serum group,high,medium and low dosage drug containing Naotong soup group. The expression of BDNF mRNA in rat hippocampal neurons after hypoxia 24 h and reoxygenation 2 h was detected in real-time quantitative PCR. Results: Real-time fluorescent quantitative PCR indicated that comparing with normal cell group,the relative expression levels of BDNF mRNA decreased significantly in model group( P〈0. 05). Comparing with the model group,The relative expression levels of BDNF mRNA increased significantly in Naotong soup groups,meanwhile high and medium doses of Naotong soup groups were significantly higher( P〈0. 05). However,normal serum group and the model group showed no significant difference comparing BDNF mRNA expression trend( P〉0. 05). Conclusions: Naotong soup can improve the expression of BDNF mRNA in rat hippocampal neurons after hypoxia-reoxygenation.
