摘要
目的观察益气化瘀方对缺血-再灌注(I/R)大鼠心肌细胞凋亡相关蛋白Bcl-2、Bax、Caspase-3、Fas及P53表达的影响。方法采用结扎左冠状动脉前降支,心肌缺血30 min,再灌注2、4 h制作缺血-再灌注模型。将大鼠随机分为7组,即假手术组、模型2 h组(缺血-再灌注2 h)、模型4 h组(缺血-再灌注4 h组)、通心络2 h组(缺血-再灌注2 h)、通心络4 h组(缺血-再灌注4 h)、益气化瘀方2 h组(缺血-再灌注2 h)、益气化瘀方4 h组(缺血-再灌注4 h)。造模前连续灌胃相应药物1周。免疫组化法测定Bcl-2、Bax、Caspase-3、Fas、P53蛋白表达。结果益气化瘀方可提高缺血-再灌注大鼠心肌Bcl-2蛋白表达量(P<0.05)、降低缺血-再灌注大鼠心肌Bax、Caspase-3、Fas、P53蛋白表达量(P<0.05)。结论益气化瘀方可减轻大鼠心肌缺血-再灌注细胞凋亡,其机理可能与上调Bcl-2蛋白表达和下调Bax、Caspase-3、fas、P53蛋白表达有关。
Objective: To investigate the Yiqi Huayu Decoction on myocardial cell apoptosis and Bcl-2,Bax,Caspase-3,fas and P53 protein expression in experimental rats with myocardial ischemia and reperfusion(ischemia/reperfusion,I/R)and to analyze the pathological damage in myocardial tissue. Methods: The myocardialischemia-reperfusion injury model was established by occluding the left anterior descending branch of coronary artery for 30 minutes and removing the ligation later to reperfusion for 2 hours,4 hours. Forty-two rats were randomly divided into seven groups: the normal(Normal) group,the sham operation(Sham) group,the saline ischemia-reperfusion 2 hours(NS-2 h) group,the saline ischemia-reperfusion 4 hours(NS-4 h) group,Tongxinluo control 2 hours group(TXL-2 h),Tongxinluo control 4 hours group(TXL-4 h),Yiqi Huayu Decoction treatment2 hours group(YQ-2 h),and Yiqi Huayu Decoction treatment 4 hours group(YQ-4 h). Bcl-2,Bax Caspase-3,Fas and P53 protein expression were tested by immunohistochemical staining method. Results: Yiqi Huayu Decoction increased the Bcl-2 protein expression(P〈0.05) and reduced the Bax,Caspase-3,Fas and P53 protein expression(P〈0.05) in rats with myocardial ischemia. Conclusion: The mechanism of Yiqi Huayu Decoction against myocardial apoptosis may be related to the increase of Bcl-2 protein expression and decrease of Bax,Caspase-3,Fas and P53 protein expression.
