高效液相色谱法测定加味葛根片中葛根素的含量

The Content of Puerarin Determined by High Performance Liquid Chromatography (HPLC) in Mixed Gegen Tablet

目的 :测定加味葛根片中葛根素的含量。方法 :反相高效液相色谱法 ,色谱条件 Hypersil ODS2 5 μm,2 0 0 mm× 4.0mm色谱柱 (大连依利特物所提供 ) ,流动相为甲醇 -水 (2 5∶ 75 ) ,流速 1.0 ml/ min,检测波长 2 5 0 nm ,柱温为室温。结果 :葛根素的进样量在 0 .10～ 0 .6 0μg范围内有良好的线性关系 ,r=0 .9998,加样回收率为 97.5 6 %(RSD=1.0 1%,n=3) ,精密度 RSD=1.0 6 %(n=5 ) ,重复性 RSD=0 .6 5 %(n=5 )。结论 :该法具有提取完全 ,操作简单快捷 ,分离度好 ,样品回收率高 ,重复性好等优点 ,是一种简便有效的检测方法。

Objective:To assay the content of puerarin in Mixed Puerarin Tablet.Methods:Reversed phrase HPLC, chromatographic conditions adopted were:hypersil ODS2 5 μm; chromatographic column, 200 mm×4.0 mm; mobile phrase, methanol-water(25∶75); flow rate, 1.0 ml/min; detection wavelength, 250 nm;column temperature, room temperature.Results:The linear relationship is good in the range of 0.10～0.60 μg content of Mixed Puerarin (r=0.999 8); the recovery rate is 97.56% (RSD=1.01%, n=3); the precision RSD=1.06% (n=5) and the repeatability RSD=0.65% (n=5).Conclusion:This method is simple, effective and repeatable for determination of puerarin in Mixed Gegen Tablet.
