hTERT和E2F-1蛋白在结直肠癌组织中的表达及其临床意义

Expressions of h TERT and E2F-1 proteins in colorectal carcinoma tissues and their clinical significance

摘要 目的 :探讨人端粒酶逆转录酶(human telomerase reverse transcriptase,h TERT)和转录因子E2F-1在结直肠癌组织中的表达及其临床意义。方法 :应用免疫组织化学法检测134例结直肠癌、74例结直肠腺瘤和28例正常结直肠黏膜组织中h TERT和E2F-1蛋白的表达,分析h TERT和E2F-1蛋白的表达与结直肠癌患者临床病理特征及预后之间的关系。结果 :h TERT蛋白在结直肠癌、结直肠腺瘤和正常结直肠黏膜组织中的阳性表达率分别为47.0%、40.5%和28.6%(P>0.05),h TERT在结直肠癌组织中的表达与Dukes分期和淋巴结转移有关(P<0.05)。E2F-1蛋白在结直肠癌、结直肠腺瘤和正常结直肠黏膜组织中的阳性表达率分别为72.4%、95.9%和100.0%(P<0.05),E2F-1在结直肠癌组织中的表达与Dukes分期和淋巴结转移有关(P<0.05)。h TERT阳性组中,Dukes C+D期患者化疗后生存率明显提高(P<0.05)。Dukes分期和h TERT蛋白表达是影响结直肠癌患者的独立预后因素(P<0.05)。结论 :h TERT和E2F-1蛋白表达与结直肠癌患者的疾病进展及预后有关,E2F-1可能参与了结直肠癌的发生,Dukes C+D期结直肠癌患者进行h TERT检测对手术后化疗有一定的指导意义。 Objective: To investigate the expressions of human telomerase reverse transcriptase (hTERT) and E2F-1 proteins in colorectal carcinoma tissues and their clinical significance. Methods: The expressions of hTERT and E2F-1 specimens of colorectal carcinoma tissues, 74 speci adenoma tissues and 28 specimens of normal c proteins in 134 mens of colorectal olo tissues were detected by immunohistochemistry. The expressions of hTERT and E2F-1 proteins with clinica rectal mucosa correlations of characteristics and prognosis of patients with colorectal carcinoma were analyzed Results: The positive rates of hTERT protein expression in colorectal carcinoma, colorectal adenoma and normal colorectal mucosa tissues were 47.0%, 40.5% and 28.6%, respectively (P 〉 0.05). The expression of hTERT in colorectal carcinoma tissues was correlated with Dukes stage and lymphatic metastasis (both P 〈 0.05). The positive rates of E2F-1 protein expression in colorectal carcinoma, colorectal adenoma and normal colorectal mucosa tissues were 72.4%, 95.9% and 100.0%, respectively (P 〈 0.05). The expression of E2F-1 protein in colorectal carcinoma tissues was correlated with Dukes stage and lymphatic metastasis (both P 〈 0.05). For colorectal cancer patients with positive expression of hTERT protein, the survival of the patients with Dukes stages C and D after chemotherapy was improved (P 〈 0.05). Dukes stage and the expression of hTERT protein were independent prognostic factors for colorectal cancer patients (both P 〈 0.05). Conclusion: The expressions of hTERT and E2F-1 proteins are closely related to the progression and prognosis of patiens with colorectal carcinoma. E2F-1 may play a role in tumorigenesis of colorectal cancer. Detection of hTERT protein may be helpful to determine post-operative chemotherapy for patients with Dukes stages C and D colorectal carcinoma.
