摘要
【目的】对异小杆线虫NLK-1大量增值技术进行研究,筛选出活性较高、毒力较好的的异小杆线虫NLK-1。【方法】采用以当地容易获得的黄粉虫作为寄主昆虫的White trap法,以线虫-共生细菌系统建立的液体繁殖法和固体繁殖法大量培养异小杆线虫NLK-1。通过评价繁殖线虫的产量以及质量,得出一种经济而高效的方法。【结果】White trap法以黄粉虫为寄主昆虫,进行活体繁殖线虫,用线虫收集盘收集线虫,收获的线虫侵染率为92.6%,产量为2.47×105IJS/g;液体培养采用PSYG培养基和TSYS培养基培养线虫,在第11~25 d内收获线虫,其中,TSYS培养基生产线虫较多,第17 d为收获最佳时间,收获的线虫侵染率为77.8%,产量为1.52×105IJS/m L;固体海绵PSYG培养基和TSYS培养基在第11~25 d内收获线虫,PSYG培养基生产线虫较多,第23 d为收获最佳时间,收获线虫侵染率为81.5%,产量为1.8×105IJS/m L。线虫侵染寄主能力White trap法最强,液体繁殖法其次,固体繁殖法略差。【结论】White trap法适宜于小批量生产;当市场需求量较大时,离体繁殖法更适宜,其中液体繁殖法更具发展潜力。
【Objective】To research mass production technology for heterorhabditis NLK- 1,and choose a heterorhabditis NLK- 1 that has higher activity and better infectivity.【Method】White trap method that took the tenebrio as the host insects was easy access to local people,and liquid propagation method and solid propagation method established by nematodes- symbiotic bacteria system were used to cultivate heterorhkabditis NLK- 1. The purpose was to find out an economic and efficient method by evaluating the yield and quality of production methods. 【Result】White trap method with tenebrio as host insects,for in vivo propagation,collecting nematode by nematodes collecting plate and harvest nematodes,infection rate was 92. 6% and production was 2. 47 × 10^5 IJS / g; Liquid Production technology using PSYG medium and TSYS medium nematodes to culture nematodes,harvest nematodes within 11- 25 d,the production of nematodes TSYS medium was more,17 d as the best time to harvest,infection rate was 77. 8% and production was 1. 52 × 10^5 IJS / ml; Solid sponge production technology using PSYG medium and TSYS medium nematodes to culture nematode,harvest nematodes within 11- 25 d,the production of nematodes PSYG medium was more,23 d as the best time to harvest,infection rate was 81. 5% and production was 1. 8 × 10^5 IJS / ml. The strongest nematodes ability to infect the host was the white trap method,the second liquid was propagation method,and the third one was solid propagation method. 【Conclusion】White trap method is suitable for small batch production; when the market was in great demand,in vitro propagation method was more suitable,in which the liquid propagation method was more potential for development.
