摘要
目的通过构建大鼠非酒精性脂肪性肝病(NAFLD)模型,观察其外周血中辅助性T细胞17(Th17)细胞频率变化及祛脂愈肝颗粒对NAFLD模型大鼠给药后的Th17细胞表达情况,探讨Th17细胞在NAFLD发病机制中及中药治疗过程中的免疫学作用。方法 SD大鼠60只,随机分为空白对照、模型对照、祛脂愈肝颗粒低/中/高剂量治疗、水飞蓟素治疗组,以高脂饮食构建NAFLD,取外周血以流式细胞术检测Th17细胞频率,进行统计分析。结果 NAFLD模型大鼠Th17细胞表达水平较正常对照组明显增高(P<0.05),祛脂愈肝颗粒中剂量组、高剂量组Th17细胞表达水平较模型组显著降低(P<0.05)。结论 NAFLD模型大鼠外周血中Th17细胞表达水平增高,祛脂愈肝颗粒能够降低NAFLD模型大鼠外周血Th17细胞的表达。
Objective To observe the frequency changes of the T helper cells 17(Th17)cells in peripheral blood and Quzhiyugan granule's therapeutic effect on NAFLD model rats' Th17 cells expression,rats model of nonalcoholic fatty liver disease(NAFLD)were made to explore Th17cells' effects in the pathogenesis of NAFLD and the immunological function of Chinese medicine during treatment.Method 60 SD rats were randomly divided into blank control group,model control group,Quzhiyugan granule low/medium/high dose treatment group,silymarin treatment group.The rats were fed with high fat diet to be rats with nonalcoholic fatty liver disease.The Th17cells' frequency in Peripheral blood was tested by flow cytometry analysis.Result The expression level of Th17 cells in NAFLD model group were significantly higher than normal control(P〈0.01).The middle and the high dose Quzhiyugan granule guoup's Th17 cellular expression were similar,showing an obvious decrease than the model group(P〈0.05).(P〈0.05).Conclusion Th17 cellular expression level is highly on NAFLD model,and Quzhiyugan granule can reduce NAFLD model of rat peripheral blood Th17 cellular.
