蒲公英绿原酸HPLC检测及不同生境对其含量的影响 被引量:1

DETERMINATION OF CHLOROGENIC ACID OF Taraxacum officnala IN DIFFERENT HABITAT BY HIGH PERFORMACE LIQUID CHROMATOGRAPHY

摘要 利用HPLC法比较分析旱生与湿生环境下蒲公英全草及各器官绿原酸含量差异,探讨影响植物生长和次生物质代谢的主要因子--生境对蒲公英绿原酸含量的影响,为进一步利用蒲公英和调节绿原酸代谢提供依据。色谱条件为Inertsil ODS-SP色谱柱(150mm×4.6,5μm);流动相为甲醇-0.4%磷酸水溶液(15:85);检测波长326nm;柱温35℃;流速1m L/min;进样量10μL。检测结果表明,旱生蒲公英全草及各器官的绿原酸含量明显高于湿生蒲公英,其全草的绿原酸含量相差近8倍。不同器官中旱生蒲公英叶的绿原酸含量最高,湿生蒲公英根的绿原酸含量最高。 The HPLC method was used to determine the chlorogenic acid( CA) contents in the whole plant and different organs of Taraxacum officnala from the wet and xeric environment. The influence of the habitat was discussed to provide theoretical basis for the applying of T. officnala and the regulating of CA metabolism. The separation was performed in Inertsil ODS-SP column( 150mm× 4.6,5μm) with methanol-0.4% phosphoric acid( 15: 85) as mobile phase. The detection wavelength was 326 nm. The column temperature was 35 ℃. The flow rate was 1.0m L / min.The injection volume was 10μL. In addition,the CA content of the whole plant and different organs of T. officnala which grew in drought was significantly higher than in wet. Eight times difference of the CA content was found between two kinds of habitat plants. Moreover,the CA content in leaves of T. officnala which living in the xeric environments was highest,but the most was found in roots in the wet environments plants.
