慢性炎症状态对强直性脊柱炎外周CD8^+记忆性T细胞的影响 被引量:1

Effect of chronic inflammation status on the peripheral memory CD8^+T cells in ankylosing spondilitis

摘要 本研究通过分析不同组别强直性脊柱炎患者外周记忆性CD8^+T细胞比例、亚群、IFN-γ,IL-2分泌及抗原特异性记忆性T细胞比例与正常对照的差异,探讨自身免疫性炎症反应对强直性脊柱炎患者记忆性T细胞的影响;主要通过流式细胞术检测强直性脊柱炎患者外周血CD8^+记忆性T细胞分布和分泌IFN-γ、IL-2的记忆性T细胞百分比及产生IFN-γ的抗原特异性记忆性T细胞百分比。结果发现与正常对照组相比,强直性脊柱炎各组患者外周总CD8^+记忆性T细胞的比例无显著差异,但亚群比例分析显示疾病组中央型记忆性T细胞比例升高,效应型记忆性T细胞比例有所下降。进一步研究发现各组强直性脊柱炎患者分泌IFN-γ的外周CD8^+记忆性T细胞比例与正常对照相比显著下降(P<0.05),而IL-2的分泌细胞比例无统计学差异;并且疾病组抗原特异性记忆性T细胞比例较正常对照明显升高(P<0.05)。因此强直性脊柱炎患者在疾病状态下,CD8^+TM的数量虽然没有变化,但其产生IFN-γ的功能下降,且抗原特异性记忆性T细胞的比例升高,这可能与患者感染高发相关。 Ankylosing spondylitis patients are prone to infection, which may relate to altered subsets and function of peripheral CD8+ memory T cells.. In this study, we set to explore the effect of autoimmune inflammation on memory T cells, we ana- lyzed the percentage of total CD8+ memory T cell and subsets profile, cytokine IL-2 and IFN-γsecreting memory cells, and per- centage of antigen-specific memory T cells in peripheral blood mononuclear cells of ankylosing spondylitis (AS) patients. The proportion of CD8+ memory T cells, functional CD8+ memory T cells and antigen-specific memory T cell were detected by flow cytometry. Compared with the healthy controls, all AS patients showed no significant difference in the proportion of total CD8+ memory T cells. However, the proportion of CD8+ central memory T cells was increased and the proportion of CD8+ ef-lector memory T cell was decreased in the patients compared with the healthy controls. Further functional study showed that IFN-γ secretion by CD8+ memory T cell were declined in all patients compare to the healthy controls(P〈0.05) ,while IL-2 se- cretion level showed no significant difference. Furthermore, percentage of antigen-specific memory T cell in the patients was in- creased compared to that in the normal controls(P〈0.05). The capability of IFN-γ secretion of CD8+ memory T cells was de- clined,and antigen-specific memory T cells was increased in the patients, which might related to the high incidence of infection in patients.
