稀释液中添加番茄红素对鸡精子保护作用的研究 被引量:2

Study on the protective effect of the lycopene added into the diluent on chicken sperm

摘要 为了研发新的鸡精液稀释液,试验在YHFB鸡精液稀释液(分别为B1液、B2液、B3液、B4液)中添加浓度分别为0,0.1,0.2,0.05 mg/mL抗氧化剂番茄红素,按1∶1的比例对鸡混合鲜精进行稀释,稀释后置于长时间保存的低温条件(5℃左右)下,定期检查精子活力,当小于或等于0.3时停止检查;置于高温恶劣环境(41℃水浴)下,每30 min检查1次精子活力,直到精子全部死亡。结果表明:低温条件下不同时间精子活力B1、B4液,B2、B3液均差异不显著(P>0.05),添加0.1 mg/mL番茄红素的B2液在48小时前精子活力稍大于B3液,48小时后B3液精子活力稍大于B2液;高温条件下不同时间精子活力各组之间无显著差异(P>0.05)。说明在低温条件下鸡精液稀释液中添加0.1 mg/mL番茄红素,能显著延长精子活力,但在41℃的高温条件下对精子的保护作用不明显。 To develop a new chicken semen diluent, the antioxidant lycopene with concentrations of 0, O. 1,0.2, and 0.05 mg / mL was added in YHFB chicken semen diluents to dilute 1/1 ratio of chicken mixed fresh semen, and named diluent B1 to diluent Bd, respectively. The diluted semen was placed under long- term storage at low temperature (5 ℃ or so) after dilution, sperm motility was checked periodically, until the sperm viability was less than or equal to 0.3. The diluted semen was put in the harsh environment of high temperature with a water bath at 41 ℃, and the sperm motility was checked once every 30 min until the sperm was all dead. The results showed that the sperm motility of diluent B1 and diluent B4 had no significant difference with that of diluent B2 and diluent B3 at different time under the low - temperature condition. The sperm motility of diluent B2 with the addition of 0.1 mg / mL of lycopene was slightly greater than that of diluent B3 before 48 h, and the sperm motility of diluent B3 was slightly greater than that of diluent B2 after 48 h. There was no significant difference( P 〉 0.05 )in the sperm motility among the 4 groups at different time under the high - temperature condition. The results indicate that the addition of 0.1 mg/mL of lycopene in chicken semen diluent can significantly prolong sperm motility under the low - temperature condition, but its protective effect in the sperm is not obvious under the high- temperature condition of 41 ℃.
