呋塞米联合右旋糖酐对罗格列酮治疗糖尿病兔的影响 被引量:3

Influence of furosemide combined with dextran on rosiglitazone in the treatment of diabetic rabbit

摘要 目的观察呋塞米联合右旋糖酐对罗格列酮治疗糖尿病兔的影响。方法将18只糖尿病兔按体重随机分为3组:对照组、呋塞米组(呋塞米0.3mg·kg-1)、呋塞米联合右旋糖酐组(呋塞米0.3 mg·kg-1+右旋糖酐5 m L·kg-1)。3组均灌服罗格列酮0.5 mg·kg-1,每天1次,连续给药3周。酶联免疫法测胰岛素(INS)、血管紧张素Ⅱ(AngⅡ)水平,硝酸还原酶法测一氧化氮(NO)水平,检测空腹血糖(FPG);用稳态模型评估法测定胰岛素抵抗指数(HOMA-IR)水平和体重。结果与给药前相比,给药后呋塞米联合右旋糖酐组FPG水平无明显变化而体重明显下降(P<0.01),对照组、呋塞米组体重无明显变化而FPG水平明显下降(P<0.01)。给药后与对照组相比,呋塞米联合右旋糖酐组NO水平明显降低(P<0.01);给药后,3组FPG、INS、HOMA-IR、AngⅡ水平及体重的差异均无统计学意义(P>0.05)。结论呋塞米联合右旋糖酐影响罗格列酮的疗效,而呋塞米不影响。 Objective To observe the impact of furosemide joint dextran on rosiglitazone in the treatment of diabetic rabbit. Methods Divide 18 diabetic rabbits into control group, furosemide group (furosemide 0. 3 mg·kg^-1), and furosemide joint dextran group (furosemide 0. 3 mg ·kg^-1 and dextran 5 mL ·kg^-1 ). Three groups were given intragas- trical administration of rosiglitazone 0. 5 mg ·kg^-1 for three consecutive weeks, once a day. Insulin (INS), angiotensin 11 (Ang II ) were tes- ted by Enzyme -linked immunoassay. Nitric oxide (NO) was tested by nitrate reductive enzymatic. Fasting plasma glucose (FPG), homeostasis model of assessment for insulin resistence index (HOMA - IR ) and weight were tested. Results Compared with that before treatment, FPG level of furosemide joint dextran group had no significant change (P 〉 0.05 ), but weight decreased obviously ( P 〈 0. 01 ). FPG levels of the control group and furosemide group significantly decreased after the treatment (P〈0.01), but weight of two groups had no significant change (P 〉 0. 05 ). Compared with the control group, NO level of furo- semide joint dextran group reduced after the treatment (P 〈0. 01 ), while NO level of furosemide group had no statistical difference after treatment (P 〉 0. 05 ). After the treatment, the levels of FPG, INS, HOMA - IR, Ang II and body weights of three groups had no statistical difference (P 〉 0. 05 ). Conclusion The combination of furosemide and dextran affected the curative effect of rosiglitazone, but furosemide does not.
