摘要
目的探讨依赖还原型辅酶I/II醌氧化还原酶1(NQO1)基因C609T多态性与桂林地区女性乳腺癌遗传易感性及临床病理特征的关系。方法采用高通量Taq Man MGB实时荧光定量PCR技术检测295例乳腺癌患者和232例健康人群NQO1基因C609T rs1800566位点的多态性分布,分析其与乳腺癌易感性及临床病理特征的关系。结果 NQO1三种基因型在两组间分布频率差异有统计学意义(P<0.05);与携带C等位基因个体相比,T等位基因携带者乳腺癌发病风险增高(P<0.05);携带CT、TT基因型的乳腺癌患者ER、PR的阳性率及淋巴结转移率高于CC基因型携带者(P<0.05)。结论 NQO1基因C609T多态性增加了乳腺癌的发病风险,并且与ER、PR、淋巴结转移有关,有望成为评价乳腺癌预后的潜在指标。
Purpose To investigate the relationship between NQO1 C609T polymorphisms and genetic susceptibilities and chnicopathological characteristics of breast cancer in Guilin. Methods The polymorphisms of NQO1 C609T rs1800566 from 295 samples of breast cancer patients and 232 samples of healthy controls were analyzed by using TaqMan MGB fluorescence quantitative polymerase chain reaction technology. The relationship between NQO1 C609T polymorphism and susceptibility and clinicopathologic parameters was also analysed. Results The frequencies of the three genotypes of NQO1 showed a significant difference in the distribution of the genotypes between patients with breast cancer and health controls (P 〈 0. 05). The risk of breast cancer was higher in T allele carriers compared with C allele carriers (P 〈0.05). The positive rate of ER, PR and the rate of lymph nodes metastasis in breast cancer with CT, TT genotype was higher than those with CC genotype (P 〈 0. 05). Conclution NQO1 C609T polymorphisms may increase breast cancer risk and correlate to ER, PR, lymph nodes metastasis, which suggest NQO1 C609T polymorphisms may be a potential biomarker for poor prognostic evaluation of breast cancers.
