双合汤对酒精性股骨头坏死兔微循环的影响 被引量:4

摘要 目的观察双合汤对酒精性股骨头坏死(AOFH)兔微循环的影响。方法将60只中国白兔随机分成正常组、模型组、美多巴组及双合汤低、中、高剂量组。除正常组外,各组给予中国白酒(乙醇含量56%)10 ml/(kg·d)灌胃。在造模同时,各给药组灌胃给药,正常组、模型组给予生理盐水。预防性灌胃给药8周后测定血浆一氧化氮(NO)、内皮素-1(ET-1)、血清超氧化物歧化酶(SOD)及丙二醛(MDA)水平。采血后处死实验动物,观察组织病理学的变化。结果与正常组比较,模型组的NO、SOD水平显著下降(P<0.01),ET-1、MDA水平显著升高(P<0.01)。与模型组比较,双合汤中、高剂量组可显著降低ET-1、MDA水平,升高NO、SOD水平(P<0.01)。双合汤中、高剂量组的NO、ET-1、SOD、MDA水平与正常组比较,差异无统计学意义(P>0.05)。模型组的空骨陷窝率显著高于正常组(P<0.01),软骨下区血管计数显著低于正常组(P<0.01)。双合汤中、高剂量组的空骨陷窝率明显低于模型组(P<0.01),软骨下区血管计数明显高于模型组(P<0.01)。双合汤中、高剂量组的空骨陷窝率、软骨下区血管计数与正常组比较,差异无统计学意义(P>0.05)。结论双合汤能有效改善AOFH兔微循环,从而防治AOFH。 【Objective】 To observe the effect of Shuanghe Decoction on microcirculation of Chinese white rabbits with alcohol-induced osteonecrosis of the femoral head(AOFH). 【Methods】 In this study 60 Chinese white rabbits were randomly divided into normal group, model group, Madopar group, and low-, medium- and high-dose Shuanghe Decoction groups. Except the normal group, each group was lavaged with Chinese spirit(alcohol content 56%) 10 ml/(kg·d). Meanwhile each dose group was lavaged with the drug. The normal group and the model group were given normal saline. After 8 weeks the rabbits were sacrificed to collect blood for determination of nitric oxide(NO), endothelin-1(ET-1), superoxide dismutase(SOD) and malondialdehyde(MDA). The rabbits were killed after collection of blood, and histopathological changes were detected.【Results】Compared with the normal group, the levels of NO and SOD in the model group were significantly decreased(P 〈 0.01), and the levels of ET-1 and MDA in the model group were significantly increased(P 〈0.01). Compared with the model group, the levels of ET-1 and MDA were significantly decreased in the medium-and high-dose of Shuanghe Decoction groups, and the levels of NO and SOD were significantly increased(P 〈 0.01). Differences in the levels of NO, ET-1, SOD and MDA between the medium- and highdose of Shuanghe Decoction groups and the normal group had no statistical significance(P 〉 0.05). Compared with the normal group, the empty bone lacuna rate in the model group was significantly increased(P 〈 0.01),and the blood vessel count in hypotegum of cartilage was significantly decreased(P 〈 0.01). Compared with the model group, the empty bone lacuna rate was significantly decreased in the medium- and high-dose of Shuanghe Decoction groups(P 〈 0.01), and the blood vessel count in hypotegum of cartilage was significantly increased(P 〈 0.01). Differences of the empty bone lacuna rate and blood vessel count in hypotegum of cartilage between the medium- and high-dose of Shuanghe Decoction groups and the normal group had no statistical significance(P 〉 0.05). 【Conclusion】 Shuanghe Decoction can effectively improve the microcirculation of rabbits with AOFH, thus prevent and cure AOFH.
