摘要
目的 探讨DACT1在乳腺癌及癌旁组织中的表达及与其他临床病理特征的相关性,并分析其与术后生存数据的关系.方法 检测120例乳腺癌组织及68例癌旁组织中DACT1的表达,比较两组表达率的差异,以及实验组DACT1表达水平与其他临床病理指标的相关性.结果 乳腺癌DACT1的阳性表达率(18.3%)显著低于癌旁组织(56.2%,P<0.05).相关分析结果显示,DACT1表达水平与肿瘤大小、淋巴结转移、临床分期、组织学分级、雌激素受体(ER)、人类表皮生长因子受体2(Her-2)存在明显相关(P<0.05);但与年龄及孕激素受体(PR)表达无明显相关(P>0.05).生存分析显示,DACT1蛋白表达水平高的乳腺癌患者预后较DACT1低表达水平的患者好(P<0.05).结论 DACT1在乳腺癌组织中的表达较癌旁组织中明显降低,且随着肿瘤大小的增大、淋巴结转移的出现、组织学分级及临床分期的增高而表达下降,并与ER表达呈正相关,与Her-2表达呈负相关,其高表达的患者预后较好,表明其可能在乳腺癌的发生发展中发挥重要作用.
Objective To detect the expression of dapper, antagonist of beta-catenin, homolog 1 Xenopus laevis (DACT1) in breast cancer and adjacent paracancerous tissues, and analyze the correlation between its expression level and other clinical features of breast cancer.To evaluate its significance in prognosis.Methods The expression of DACT1 was detected by immuneohistochemical method in 120 breast cancer patients and 68 adjacent paracancerous tissues.The difference between the two groups and the relationship between DACT1 expression and other clinical features were tested.Results The expression of DACT1 in breast cancer group (18.3%) was lower than adjacent paracancerous tissues group (56.2%, P 〈0.05).DACT1 expression level showed positive correlation with tumor size, lymph node metastasis, clinical stage, histological grade, estrogen receptor (ER) and human epidermalgrowth factor receptor-2 (Her-2) expression, but no statistical correlation with age and progesterone receptor (PR) expression.Statistical analysis showed that compared with patients with low expression of DACT1, patients with high DACT1 expression level had significantly good prognosis, and the difference had statistical significance (P 〈 0.05).Conclusion The expression of DACT1 is obviously declined in breast cancer patients than that in adjacent paracancerous tissues, and its expression was correlated with clinical stage.The higher expression of DACT1 indicates the good survival rate.These result may implicate that DACT1 is important in the occurrence and development of breast cancer.
