不同生境下茶树内生真菌多样性分析及其对PDA培养基pH值的影响 被引量:3

摘要 本文以茶树无性系品种龙井茶叶为材料,结合对真菌ITS区段的分子鉴定,首次对茶树内生真菌在不同生境、不同季节的种类与多样性进行了研究,并对茶树内生真菌的泌酸性进行了初步的分析。结果表明:林下茶园相对于非林下茶园有着更好的物种多样性和更加良好的物种丰富度,但两者的均匀度指数相近,两者的种群也处于极高相似度水平。不同的内生真菌对PDA液态培养基的p H值有不同的影响。大多数真菌在培养的过程中会加重培养基的酸化,但胶孢炭疽菌的菌株JF04、Phomopsis sp2.的菌株EF3和EF6、尖孢镰刀菌的菌株JZ02、球毛壳菌的菌株EF9、串珠状赤霉的菌株ZH01以及围小从壳菌的菌株EF5能较大提高p H值,值得深入研究。 Longjingchaye,clonal tea cultivars has been chosen as material in this study. With molecular identification of fungal ITS(internal transcribed spacer)section,we firstly conducted a systematic study on species and diversity of endophytic fungi fromCamelliasinensisamong different habitats and seasons. Meanwhile a preliminary analysis of endophytic fungi's acid-secretion has been done in this study. The results show that compared with naked tea garden, under-forest tea garden has a better diversity and a better richness of species. But there are similar Evenness index and Shannon-Wiener index. Different endophytic fungi have different effects on pH value of liquid PDA medium. Most fungi will increase the process of medium’s acidification during the culture,but compare with CK,fungal strains contain JF04(Colletotrichumgloeosporioides),EF3 and EF6(Phomopsissp2.),JZ02(Fusariumoxysporum),EF9 (Chaetomiumglobosum),ZH01(Gibberellamoniliformis),and EF5(Glomerellacingulata),can greatly increase medium’s pH value which should be noticed and should be studied in depth.
