摘要
目的与方法 :用聚合酶链式反应 单链构象多态性分析 (PCR SSCP)方法观察α粒子诱发人支气管上皮细胞系BEP2D细胞转化过程中相关基因的改变。结果 :细胞转化过程中 ,双链断裂修复基因XRCC5发生碱基突变 ,改变从转化早期过程就持续存在 ;而p16基因exon2和hMSH2基因exon12未见变化。结论 :XRCC5基因在α粒子照射后早期发生突变 ,使细胞丧失修复双链断裂损伤的能力 ,是α粒子诱发支气管上皮细胞转化过程中的启动事件之一。
Purpose and Methods: To detect related genes mutation in human bronchial epithelial(BEP2D) cells transformed by α-particles and by using PCR amplification and single-strand conformation polymorphism(SSCP). Results: The point mutation of double-strand break repair gene XRCC5 occurred at the early stage of transformation, but no mutation was found in the exon 2 of p16 gene and exon12 of hMSH2 gene. Conclusion: Mutated XRCC5 gene can't repair DNA lesions induced by α-particles, and plays as one initiator in the process of transformation induced by α-particles.
