摘要
目的建立SD大鼠与Wistar大鼠C6胶质瘤模型,通过对比成瘤率及观察肿瘤生长特性从而判断大鼠种类的选择及性别的选择对实验的影响。方法大鼠c6胶质瘤细胞进行体外培养,应用立体定向技术,将含有1×106个细胞的DMEM高糖溶液20肛L注入大鼠右侧尾状核区。接种后分别对SD大鼠及Wistar大鼠模型进行一般观察和MR检查,并取组织用HE染色法及透射电镜观察肿瘤的存在。结果雄性SD与Wistar大鼠的成瘤率均为100%,且均无远处转移,而雌性组成瘤率不足100%;但SD大鼠的成瘤时间较长,且部分SD大鼠肿瘤有自愈倾向,而Wistar大鼠组无类似情况。结论Wistar大鼠胶质瘤模型性状更接近人脑胶质瘤,更适合应用于脑胶质瘤的研究,SD大鼠可用于需要动物长期生存的实验研究,雌性大鼠可做为雌性对照参与实验。
OBJECTIVE To establish a SD rats and Wistar rat C6 glioma model, by comparing the tumor rate and observe the tumor growth characteris to determine the rat species selection and the choice of gender affect the experi- ment. METHODS Rat C6 glioma vitro, the application of stereotactic technique, 1X 106 ceils will contain 20 mu L DMEM sugar solution into the right caudate nucleus in rats. After inoculation of SD rats and Wistar rats model of gen- eral observation and MRI, and take the organization with HE staining and transmission electron microscope the existence of the tumor. RESULTS The male SD and Wistar rat tumor rate were 100%, and no distant metastasis, while the female of tumor rate less than 100% ; But SD rats into tumor for a long time, and part of SD rat tumor has a tendency to heal, and Wistar rats group no similar conditions. CONCLUSION Wistar rat glioma model properties is closer to the human brain glioma, more suitable for application in the research of glioma, SD rats can be used to need animal experimental study of the long-term survival of, female rats as the female controls to participate in the experiment.
