摘要
目的观察和分析对胃癌患者血清中肿瘤相关基因超甲基化进行检测的可行性与临床意义。方法收集2014—03—2015-02我院原发性胃癌患者40例进行临床研究,并将其作为研究组。同时选同期我院健康体检的40例健康人群作为对照组。采用甲基化特异性PCR(MSP)检测方法对两组患者的E—cadherin、GSTP1、hMLH1、p15和p16等基因的启动子超甲基化进行检测,并进行比较分析。结果E-cadherin、GSTP1、hMLHI、p15和p16等基因在胃癌组织中的超甲基化率依次为77.5%、22.5%、30.0%、60.0%和70.0%,血清中依次为37.5%、15.0%、12.5%、25.0%和30.0%。34例患者的血清中可检测到异常甲基化。结论胃癌患者的血清中能够同时检测到多个基因的异常甲基化,血清肿瘤相关基因的超甲基化检测存在较高的敏感度和特异性,具有良好的临床检测意义。
OBJECTIVE To observe and analyze the feasibility and clinical significance of the detection of tumor as- sociated gene methylation in serum of patients with gastric cancer. METHODS The collected 2014 March to 2015 Feb 2008 in our hospital admissions from primary gastric carcinoma in 40 patients for clinical research and as the study group. At the same time over the same period in our hospital 40 cases of healthy healthy people as control group were studied. Methylation specific PCR (MSP) method was used to detect the promoter methy|ation of eadherin, GSTP1, hMLH1, p15, p16, E-and other genes in the two groups, and to compare and analyze. RESULTS Cadherin GSTP1, hMLH1, p15, p16, E- and other genes in gastric cancer tissues were 77.5~, 22.5~, 30.0~, 60.0~, 70.0%, 37.5~, 15.0%, 12.5 ~, 30.0 ~, 25.0 ~//00, respectively. Abnormal methylation can be detected in the serum of 34 patients. CONCLUSION The serum of patients with gastric cancer can detect the abnormal methylation of multiple genes, and the detection of serum tumor related genes has high sensitivity and specificity, and has good clinical detection significance.
